摘 要:目的探讨衰老标记蛋白-30和与衰老相关的β-半乳糖苷酶在老年性白内障患者的晶状体上皮细胞凋亡表达的相关性。方法 248例年龄相关性白内障患者(其中126例皮质性白内障154只眼,122例核性白内障有142眼),对每个患者进行晶状体前囊中心部的环形撕囊。TUNEL法观察晶状体上皮细胞凋亡,免疫组织化学染色和实时PCR检测蛋白与mRNA的表达水平。分析衰老标记蛋白-30和β-半乳糖苷酶蛋白表达水平与晶状体上皮细胞凋亡的关系。结果衰老标记蛋白-30在晶状体前囊中心部比晶状体前囊周边有较低的蛋白表达水平,而衰老相关的β-半乳糖苷酶的表达在中央部分明显高于周边部分。与核性白内障患者相比,皮质性白内障患者的衰老标志物蛋白-30和mRNA的表达水平降低了,而衰老相关的β-半乳糖苷酶的表达升高了。TUNEL检测结果显示,两组晶状体上皮细胞的前囊中心部凋亡率明显高于周围部分。在晶状体前囊区中心或周围,皮质性白内障比核性白内障晶状体上皮细胞凋亡明显增高。结论在核性白内障和皮质性白内障,衰老标记蛋白-30和衰老相关的β-半乳糖苷酶的表达均与晶状体上皮细胞的凋亡有相关性。Objective To investigate the correlation between the expression of senescence marker protein-30 and senescence-associated 13-galactosidase in age-related cataract patients. Methods A total of 248 age-related cataract patients ( 122 cases with nuclear cataract in 142 eyes and 126 cases of cortiealcataract with 154 eyes) were enrolled in our study. An annular tear of the central part of anterior lens capsules was performed for each patient. Immunohisto- chemical staining and real-time PCR were used to detect the protein and mRNA expression lev- els, and TUNEL was used to assess lens epithelial cells apoptosis. Comparisons of protein ex- pression levels and lens epithelial cells apoptosis were made between the two groups. Results the senescence marker protein-30 in the center of the anterior lens capsule was lower than before the lens capsule surrounding the protein expression level, and the expression of senescence-associated β-galactosidase in the central part was higher than the surrounding part. Compared with patients with nuclear cataract, the expression of senescence marker protein-30 and mRNA was decreased in the patients with cortical cataract, while the expression of senescence- associated β-galactosidase was increased. The results of TUNEL showed that the apoptosis rate of the anterior capsule in the two groups was significantly higher than that in the peripheral part. In the center or periphery of the anterior capsule of the lens, the apoptosis of cortical catar-act was higher than that of nuclear cataract. Conclusion The expression of senescence marker protein-30 and senescence-associated β-galactosidase is correlated with the apoptosis of lens epithelial cells in nuclear cataract and cortical cataract.
