摘  要:目的探索浓缩血小板制备血小板裂解液的方法并初步确定所制备血小板裂解液的质量特性。方法富血小板血浆法制备浓缩血小板,以-80℃冰冻8 h/12 h/24 h,37℃空气浴融化反复冻融制备血小板裂解液,检测裂解效率。p H计检测p H值,生化分析仪检测TP、ALB浓度,ELISA方法检测PDGF、VEGF、EGF、TGF-β、IGF-1等细胞因子浓度,并以95%参考区间确定其质量特性。结果 -80℃12 h,37℃反复冻融6次血小板裂解达到>95%,p H、TP、ALB、PDGF-AA/AB/BB、VEGF、EGF、TGF-β、IGF-1 95%参考区间分别为6.72-7.38、(45-57)g/L、(22-38)g/L、(0.98-2.01)ng/m L、(122-293)ng/m L、(5.35-15.6)ng/m L、(0.49-1.82)ng/m L、(2.97-8.03)ng/m L、(177-290)ng/m L、(95-154)ng/m L。结论建立了以浓缩血小板反复冻融制备血小板裂解液的有效方法并初步确立了其质量标准。Objective To prepare and characterize platelet lysate( PL) made from platelet concentrates( PCs). Methods PCs were prepared by platelet rich plasma method. The PL was prepared by repeated frozen at-80℃ for 8 h/12 h/24 h and thawing at 37℃ for 6 times. Lysis efficiency,p H,concentrations of TP,ALB,PDGF,VEGF,EGF,TGF-β,IGF-1 were tested and 95% reference intervals were calculated respectively. Results Over 95% of the PCs can be lysed by repeated-80℃ 12 h frozen and 37℃ thawing for 6 times. Reference intervals of p H,TP,ALB,PDGF-AA/AB/BB,VEGF,EGF,TGF-β,IGF-1 were 6. 72-7. 38,( 45-57) g/L,( 22-38) g/L,( 0. 98-2. 01) ng/m L,( 122-293) ng/m L,( 5. 35-15. 6) ng/m L,( 0. 49-1. 82) ng/m L,( 2. 97-8. 03) ng/m L,( 177-290) ng/m L,( 95-154) ng/m L,respectively. Conclusion In this study,an effective method of the preparation of PL has been established,and Preliminary characterization has been achieved.
