摘  要:目的:观察蛇床子素对人胃癌细胞BGC-823凋亡作用的影响。方法:体外培养人胃癌细胞株BGC-823,予以不同浓度蛇床子素干预,MTT法检测细胞存活情况并筛选最适作用药物浓度,Hochest 33258染色法观察细胞核形态改变,免疫印迹法检测凋亡蛋白Caspase3、Caspase 9的表达情况。结果:蛇床子素能显著抑制BGC-823细胞增殖,并诱导凋亡小体形成,蛇床子素干预细胞后能增加凋亡相关蛋白Caspase3、Caspase 9的表达水平。结论:蛇床子素能诱导人胃癌细胞BGC-823凋亡。Objective To investigate the effect of osthole on the apoptosis of human gastric cancer BGC-823 cells. Methods: Human gastric cancer BGC -823 cells were cultured in vitro and treated with different concentrations of osthole. MTT assay was used to measure cell viability and screen out the optimal drug concentration, Hochest33258 staining was used to observe the nuclear morphological changes, and Western blot was used to measure expression of caspase -3 and caspase -9. Results: Osthole significantly inhibited the proliferation of BGC-823 cells, induced the formation of apoptotic bodies, and significantly increased the expression of caspase-3 and caspase-9. Conclusions: Osthole can induce the apoptosis of human gastric cancer BGC-823 cells.
