摘  要:目的探讨染色质重塑蛋白4A(CHMP4A)及睾丸特定蛋白-2(TSPYL-2)基因在食管癌早期癌变中的作用。方法采用RT-PCR、免疫组化法从RNA及蛋白水平检测CHMP4 A及TSPYL-2在食管鳞状细胞癌组织、癌化区域组织及正常食管黏膜组织中的表达。结果 CHMP4A及TSPYL-2基因在食管鳞状细胞癌组织和正常食管黏膜组织中的表达差异有统计学意义(P<0.05),在癌化区域组织和正常食管黏膜组织中的表达差异有统计学意义(P<0.05),且CHMP4 A在正常食管黏膜、癌化区域组织、食管鳞状细胞癌组织中的表达依次增高,TSPYL-2基因在正常食管黏膜、癌化区域组织、食管鳞状细胞癌组织中的表达依次降低,CHMP4A及TSPYL-2蛋白表达的变化趋势与CHMP4A及TSPYL-2基因一致。结论差异表达基因CHMP4A及TSPYL-2可作为食管鳞状细胞癌早期基因备选基因进行后续实验。Purpose To investigate the role of CHMP4A and TSPYL-2 in early pathogenesis of esophageal cancer. Meth- ods Through comparison of the four subtractive libraries, early esophageal squamous cell carcinoma genes CHMP4A and TSPYL-2 were chosen for further study. Through RT-PCR and immunohistochemistry methods, CHMP4A and TSPYL-2' s ex- pression was detected in esophageal squamous cell carcinoma tis- sue, cancerous tissue and normal esophageal mucosa. Results CHMP4A and TSPYL-2 expression between esophageal squa- mous cell carcinoma and normal esophageal epithelium tissue had significant differences (P 〈 0. 05 ), and the CHMP4A gene expression in esophageal mucosa, field cancerization areas, e-sophageal squamous cell carcinoma tissue increased, while TSPYL-2 gene expression in esophageal mucosa, field canceriza- tion areas, esophageal squamous cell carcinoma tissue de- creased, which were consistent with the protein expression of CHMP4A and TSPYL-2. Conclusion CHMP4A and TSPYL-2 genes are differentially expressed in esophageal squamous cell carcinoma, which can be used as alternative genetic markers for further research.
