摘 要:以不同含油量的甘蓝型油菜品种或品系为材料,采用ELISA法检测了油脂累积过程中乙酰辅酶A羧化酶、葡萄糖-6-磷酸脱氢酶、磷脂酸磷酸酯酶、二酰甘油酰基转移酶等4种关键酶在不同发育时期的种子和角果皮中的酶活性变化,并分析了酶活性与种子含油量的相关性。研究结果表明,不同含油量的油菜品种在种子和角果皮中酶活性的变化趋势基本一致,在开花后第17天至第24天迅速增加,第38天时达到峰值,然后逐渐下降,但角果成熟时间较短的酶活性下降很快,如花油8号的角果成熟期只有40.2 d,比其他3个品种早熟1周。相关性分析表明种子中DGAT、PPase、G6PDH的酶活性与菜籽含油量呈极显著正相关,ACCase酶活性与菜籽含油量呈显著正相关,而角果皮中ACCase、G6PDH、PPase、DGAT的酶活性与菜籽含油量呈正相关,但没达到显著水平,其中4种酶的酶活性与菜籽含油量的相关性大小依次为DGAT、PPase、G6PDH、ACCase。因此,在进行高含油量油菜品种选育时应着重考虑油脂累积中DGAT、PPase的基因表达水平和酶活性水平。With brassica napus varieties or strains with different oil contents as experimental materials, the activity of four key enzymes in the process of oil accumulation, namely, acetyl - coa carboxylase ( accase), glucose - 6 - Phosphate dehydrogenase ( G6PDH), phospha - tidic acid phophyhydrolase ( ppase), and diacylglycero - lacyl transferase( DGAT), were detected by ELISA methods in seed and pod peel at different developmental stages, and the correlation between enzyme activity and seed oil content was analyzed. The results indicated that the variation trend of enzyme activities in seeds and pod peel of rapeseed cultivars with different oil contents was basically the same. The enzyme activity was increased rapidly from the 17th day after flowering (DAF) to the 24th DAF, and reached the peak at the 38th DAF, then it was decreased gradually. However, the enzyme activity was decreased rapidly in Huayou 8 with shorter pod ripening time of 40.2 days, earlier than the other three varieties by a week. The correlation analysis showed that the activities of DGAT, ppase and G6PDH in seeds were extremely significant positively correlated with the seed oil content, the activities of accase were positively correlated with the seed oil content, While the activity of accase, G6PDH, ppase and DGAT in pod peel was positively correlated with the oil content of rapeseed, which did not reach the significant level. The correlation levels of the four kinds of enzymes with oil content in order were as follows: DGAT, ppase, G6PDH and accase. Therefore, the gene expression level and enzyme activity level of DGAT and tent rapeseed varieties.
