片仔癀联合多西他赛通过STAT3信号通路抑制Tca8113细胞的增殖

摘　　要：【目的】探讨片仔癀(Pien Tze Huang,PZH)联合多西他赛(docetaxel,DOC)对舌鳞癌Tca8113细胞凋亡、细胞增殖的影响及可能机制。【方法】PZH与DOC联合作用Tca8113细胞,应用Hoechst 33258染色法观察细胞核形态的变化,MTT法检测细胞的增殖,Western blotting法检测两药联用对Tca8113细胞中STAT3等信号通路的影响。【结果】PZH与DOC联用可使细胞核呈现致密浓染、核固缩等现象,对Tca8113细胞增殖的抑制作用有协同作用。联合给药组与单药组相比可明显降低p-STAT3和Bcl-2蛋白的表达。【结论】PZH与DOC联合应用可通过抑制STAT3通路抑制Tca8113细胞的增殖。[ Objective ] To explore the effect of Pien Tze Huang （PZH） combined with docetaxel （DOC） on the apoptosis and proliferation of TcaS113 ceils and its related mechanism. [ Methods ] Tca8113 cells were cuhured with PZH combined with DOC. Morphological changes of cell nuclei were observed by Hoechst 33258 staining, the proliferation of TcaSll3 cells was determined by MTT assay, and the effect of the two drugs on the signal pathway was assessed by Western blotting. [ Results ] Morphological changes including chromatic agglutination and karyopyknosis were found in Tca8113 cells incubated with PZH combined with DOC. The combined treatment significantly restrained the cell proliferation and the expression of p-STAT3, Bcl-2, compared with the monotherapy. [ Conclusion ] PZH combined with DOC restrains the cell proliferation by inhibiting STAT3 signal pathway in Tca8113 cells.
