摘 要:目的探究黄芪甲苷对大鼠局灶性脑缺血/再灌注损伤的影响。方法采用改良线栓法模拟局灶性脑缺血/再灌注损伤。选取健康清洁级♂SD大鼠40只,随机分为假手术组、脑缺血/再灌注组、黄芪甲苷组和溶剂对照组。除假手术组外,其他各组缺血2 h、再灌注24 h。黄芪甲苷组于再灌注的同时腹腔注射黄芪甲苷20 mg·kg^(-1),溶剂对照组腹腔注射相同体积的溶剂。根据Zea Longa评分标准对各组大鼠进行神经功能学的评分,挑选模型成功的大鼠;TTC法检测脑梗死体积;尼氏染色法观察大鼠大脑皮质区细胞形态学的变化;透射电子显微镜观察大脑皮质区细胞超微结构的变化。结果假手术组未见神经功能缺损症状,脑梗死体积为零。与假手术组比较,其他各组均有一定的神经功能缺损症状、不同程度的神经细胞损伤,脑梗死体积增加(P<0.05);与脑缺血/再灌注组比较,黄芪甲苷组可明显改善神经功能缺损症状、减轻神经细胞损伤、减小脑梗死体积(P<0.05),而溶剂对照组无明显变化(P>0.05)。结论黄芪甲苷可减轻大鼠局灶性脑缺血/再灌注损伤,对神经细胞发挥保护性作用。Aim To investigate the effect of astragaloside Ⅳ on focal cerebral ischemia-reperfusion injury in rats.Methods The focal cerebral ischemia/reperfusion of rat left middle cerebral artery occlusion(MCAO)was induced by suture method.Male SD rats were randomly divided into sham operation group,cerebral ischemia/reperfusion group,astragaloside IV group and solvent control group.Except for the sham operation group,the others were subjected to ischemia 2h and reperfusion 24h.Then,rats with successful model were chosen for the detection of various indexes.Astragaloside Ⅳ group was injected intraperitoneally with astragaloside Ⅳ(20 mg·kg^(-1))at the same time as reperfusion,while solvent control group was injected with the same amount of solvent.TTC staining was used to detect the volume of cerebral infarction,and Nissl staining to observe the changes of histomorphology,and transmission electron microscope(TEM)to observe the ultrastructure of the cells.ResultsThere was no neurological deficit in the sham operation group,and the volume of cerebral infarction was zero.Compared with the sham operation group,there were some increased neurological deficits,nerve cell damage and cerebral infarction volume in other groups(P<0.05).Compared with the cerebral ischemia/reperfusion group,the nerve function damage could be significantly improved,the damage of neurons reduced,and the volume of cerebral infarction decreased(P<0.05)in astragaloside Ⅳ group,and there was no obvious change in the solvent control group(P>0.05).Conclusion Astragaloside Ⅳ can reduce the focal ischemia/reperfusion injury in rats and protect nerve cells from damage.
