摘  要:目的建立发病过程近似于人类2型糖尿病,且具有胰岛素抵抗特征的动物模型。方法健康雄性SD大鼠38只,随机分为对照组和模型组,模型组24只雄性SD大鼠先喂以高脂高糖饲料4周,再注射链脲佐菌素(STZ),必要时追加一次。对照组14只雄性SD大鼠用普通饲料喂养。定期监测大鼠一般情况、空腹体重及血糖,注射STZ后第四周检测血浆胰岛素水平。结果模型组大鼠较对照组表现为多饮,多食,多尿及体重减轻,血糖、胰岛素水平明显高于对照组,胰岛素敏感性下降较对照组明显。结论通过高脂高糖饮食结合STZ注射成功复制出了实验性2型糖尿病动物模型,是研究2型糖尿病及其并发症发病机制的理想动物模型。Objective To establish an animal model with an insulin resistance characteristic similar to that of human type 2 diabetes mellitus. Methods 38 healthy male SD rats were randomly divided into the control group and the model group. The model group consisted of 24 male SD Rats were fed with high fat and high sugar diet for 4 weeks,then injected with streptozotocin(STZ),and added once if necessary. The control group consisted of 14 male SD rats fed with common diet. The general condition,fasting weight and blood sugar of rats were monitored regularly,and the level of plasma insulin was detected in the 4 th week after STZ injection. Results Compared with the control group,the rats in the model group exhibited polydipsia,polydipsia,polyuria,and weight loss. The blood glucose and insulin levels were significantly higher than those in the control group,and the insulin sensitivity decreased significantly compared with the control group. Conclusion An experimental animal model of type 2 diabetes mellitus was successfully replicated by high fat and high carbohydrate diet combined with STZ injection. It is an ideal animal model for the study of the pathogenesis of type 2 diabetes mellitus and its complications.
