摘 要:目的 :观察高脂血症大鼠蓝斑和蓝斑下核多巴胺 β 羟化酶 (dopamine β hydroxylase,DβH)免疫阳性神经元的变化。方法 :用ABC免疫细胞化学方法显示DβH免疫阳性神经元。结果 :对照组大鼠各时期蓝斑和蓝斑下核DβH免疫阳性神经元的数量未见明显变化 ;高脂组大鼠 7d ,蓝斑和蓝斑下核DβH免疫阳性神经元与对照组比较无明显改变 ,但随着饲喂高脂饮食有逐渐增多的趋势 ,30d明显增多 ,特别是高脂饮食 90d后 ,DβH免疫阳性神经元的数量增多明显 ,且着色较深。蓝斑与蓝斑下核DβH免疫阳性神经元的变化具有明显的一致性 ,均随着饲喂高脂饮食时间延长而增多 ;定量分析显示 ,DβH免疫阳性神经元的数量在 30d和 90d与对照组相比均明显增多 (P <0 .0 1 ) ,同组内 30d和 90d均较 7d明显增多 (P <0 .0 1 )。结论 :脑内NE能神经系统的功能活动增强可能与动脉粥样硬化的发生有关。Objective:To observe dopamine-β-hydroxylase(DβH) immunopositive neurons of locus ceruleus and subceruleus nucleus in hyperlipidemic rats. Methods:After establishing the hyperlipidemic rat model,ABC immunocytochemical technique was used to demonstrate DβH immunopositive neurons. Results:The number of DβH immunopositive neurons in locus ceruleus and subceruleus nucleus were similar to each other in 3 different periods in control group.After taking hyperlipidemic food,the number of DβH immunopositive neurons at 7 d were no differences between the 2 groups. However,at 30 d,the number of DβH immunopositive neurons increased significantly in experimental group. Especially, at 90 d,the DβH immunopositive neurons were much more than these of 30 d hyperlipidemic group,and the dyeing of immunopositive neurons were deeper. The changes of DβH immunopositive neurons in locus ceruleus had obvious concordance with in subceruleus nucleus. Quantiative analysis suggested that the number of DβH immunopositive neurons significantly increased compared with control group,In the same hyperlipidemic group,the number at 30 d and 90 d were more than at 7 d significantly.Conclusions: The reinforce of norepinephrine system in the brain may be related to the development of atherosclerosis.
