摘 要:采用超声波辅助法提取石楠叶中的总黄酮,以双波长分光光度法测定总黄酮含量,并用DPPH法对石楠叶提取液中的总黄酮进行抗氧化活性评价。结果表明,在测定波长为418 nm,参比波长为465 nm的条件下,芦丁标准曲线回归方程为ΔA=0.025 5ρ-0.007 3,芦丁质量浓度在3.86~38.6 mg/L范围内具有良好线性关系(R^2=0.999 59),方法的检出限(3S/N)为0.27 mg/L,平均回收率为93.36%,精密度(RSD)为1.12%(n=6),总黄酮含量测定结果为2.17%;DPPH抗氧化活性测定结果表明,与常用抗氧化剂维生素C相比,石楠叶黄酮具有更强的抗氧化活性。Ultrasonic assisted method was applied to extract the total flavonoids in Photinia serrulata leaves,while the double-wavelength spectrophotometry was applied to assay the total flavonoids of the leaves.The DPPH was used to evaluate the antioxidant ability of the flavonoids in photinia serrulata extract.Results showed that in the condition of detective wavelength for 418 nm,reference wavelength for 465 nm,the rutin standard curve return equation was ΔA = 0. 025 5ρ-0. 007 3,When the mass concentration of rutin was between 3. 86 ~ 38. 6 mg/L,it had a good linear relationship( R^2= 0. 999 59),the method detection limit( 3S/N) was 0. 27 mg/L,the average recovery rate was 93. 36%,with precision( RSD) was 1. 12%( n = 6),the total flavonoids content of photinia serrulata leaves was 2. 17%; the DPPH assay result showed that photinia serrulata leaves had more antioxidant activity of flavonoids,compared with the commonly used antioxidant vitamin C.
