摘  要:目的 :了解兴奋性氨基酸在点燃癫痫模型中的改变及其作用。方法 :采用印防己毒腹腔注射制作SD鼠慢性点燃模型 ,并将其分为对照组、癫痫发作组、癫痫发作间期组和苯巴比妥钠干预组。用高效液相色谱仪测定颞区脑组织天门冬氨酸和谷氨酸浓度。结果 :癫痫发作组、癫痫发作间期组和苯巴比妥钠干预组SD鼠颞区脑组织天门冬氨酸浓度 [分别是 (0 97± 0 2 9) μmol·g- 1 ,(1 10± 0 5 4 ) μmol·g- 1 ,(1 0 7± 0 4 1) μmol·g- 1 ]较对照组SD鼠颞区脑组织天门冬氨酸浓度 [(0 6 5± 0 17) μmol·g- 1 ]明显增高 ;三组SD鼠颞区脑组织谷氨酸浓度 [分别是 (9 5 4± 2 6 3) μmol·g- 1 ,(9 5 5± 2 35 ) μmol·g- 1 ,(8 93± 1 89) μmol·g- 1 ]较对照组SD鼠颞区脑组织谷氨酸浓度 [(6 2 1± 2 0 9) μmol·g- 1 ]明显增高。结论 :在该模型中 ,兴奋性氨基酸明显增高 ,可能参与癫痫的发生 ;用苯巴比妥钠可以阻止癫痫的发生 ,但不能阻断兴奋性氨基酸增高。Objective To investigate the changes and action of excitory amino acids (EAA) in kindling models. Methods Kindling models of Sprague Dawley (SD) rats were induced by injection of picrotoxin (PTX) intraperitoneally. The SD rats were randomly divided into four groups: control group (Group A), epilepsy group in the ictal period (Group B), epilepsy group in the interictal period (Group C) and phenobarbital group (Group D). High performance liquid chromatography (HPLC) was used to determine the contents of aspartic acid (Asp) and glutamic acid (Glu) in temporal tissues of the SD rats. Results The average contents of Asp in Group B, C, and D were (0.97±0.29)μmol·g -1 , (1.10±0.54)μmol·g -1 , (1.07±0.41)μmol·g -1 respectively. The Asp contents in Group B, C, and D were obviously higher than those in Group A (0.65±0.17)μmol·g -1 . The average levels of Glu in Group B, C, and D were (9.54±2.63)μmol·g -1 , (9.55±2.35)μmol·g -1 , (8.93±1.89) respectively, which were significantly higher than those in Grouop A [(6.21±2.09)μmol·g -1 ]. Conclusion In kindling models, the levels of EAA in temporal tissues increase significantly, which take part in epileptic seizure. Phenobarbital can't prevent the increase of EAA contents.
