摘 要:目的 :观察补阳还五汤对凝血酶诱导血管内皮细胞释放NO ,vWF ,TFPI和表达TF的影响。方法 :对传代培养的新生牛主动脉内皮细胞 (4~ 8代 )分别给予不同处理 ,取上清液测NO ,vWF ,TFPI;取细胞冻融液测定TF活性。结果 :①凝血酶促进血管内皮细胞表达TF ,较对照组明显增高 (12 .86± 2 .4 3vs 4 .6 9± 0 .83,P<0 .0 1) ,呈剂量依赖性 (r =0 .985 ,P <0 .0 1,0~ 2 0U·ml- 1 ) ,并促进vWF的释放 (18.4 3± 3.2 0vs6 .4 2± 2 .84 ,P <0 .0 1) ,补阳还五汤则抑制TF表达和vWF的释放 (P <0 .0 1) ;②补阳还五汤促进血管内皮细胞释放NO ,较对照组明显升高 (5 .93± 0 .94vs 1.6 3± 0 .2 7,P <0 .0 1) ;③凝血酶能抑制血管内皮释放TFPI (0 .6 2± 0 .38vs 2 .6 4± 0 .93,P <0 .0 1) ,补阳还五汤对凝血酶的这种抑制作用没有明显的影响。结论 :补阳还五汤能抑制凝血酶诱导血管内皮细胞表达TF和释放vWF ,促进血管内皮细胞表达NO。Objective To study the effects of Buyang huanwu decoction (BHD) on the release of von Willebrand factor (vWF), nitric oxide (NO) and tissue factor pathway inhibitor (TFPI) and the expression of tissue factor (TF) induced by thrombin in cultured bovine aortic endothelial cells (BAECs). Methods BAECs from neonatal oxen were cultured and 4~8 passages were used. The cells were incubated for 8 hours after the addition of different treatments. The supernatant was used to measure TFPI, NO, vWF and BAECs lysate to determine TF activity. Results ①Compared with the controls, thrombin enhanced the expression of TF activity (12.86±2.43 vs 4.69±0.83, P<0.01) and the effects were in a dose dependent manner (r=0.985, P<0.01), BHD inhibited the effects of thrombin. ②Compared with the controls, thrombin promoted the release of vWF from the cultured endothelial cells (18.43±3.20 vs 6.42±2.84, P<0.01) and BHD inhibited the effect of thrombin (P<0.01). ③BHD increased the release of NO from BAECs. ④Thrombin inhibited the activity of TFPI, but BHD had no marked effects on the release of TFPI from BAECs. Conclusion BHD can inhibit the expression of TF and the release of vWF induced by thrombin and promote the release of NO in cultured BAECs.
