摘  要:目的探讨黄芩素对人表皮鳞癌A431细胞线粒体凋亡通路的影响。方法将人表皮鳞癌A431细胞分为空白对照组和6个黄芩素处理组,黄芩素处理组分别加入相应浓度(10μg/ml、20μg/ml、30μg/ml、40μg/ml、50μg/ml、60μg/ml)黄芩素干预,空白对照组加入等体积二甲基亚砜(DMSO)。分别于干预后24 h、48 h、72 h检测各组细胞生长抑制率。另取A431细胞分为空白对照组和3个黄芩素处理组,黄芩素处理组的细胞分别加入不同浓度(10μg/ml、20μg/ml、30μg/ml)黄芩素干预,空白对照组加入等体积DMSO,干预24 h后检测各组细胞凋亡及细胞线粒体跨膜电位的情况,细胞胞浆中细胞色素C和调亡诱导因子(AIF)水平以及细胞半胱氨酸天冬氨酸蛋白酶(Caspase)-9、Caspase-3的活性。结果干预24 h、48 h及72 h后,黄芩素对A431细胞相应的半数抑制浓度(IC50)分别为82.29μg/ml,24.67μg/ml和18.66μg/ml;10μg/ml、20μg/ml、30μg/ml黄芩素处理组总凋亡细胞率、细胞胞浆中AIF及细胞色素C含量、细胞Caspase-3和Caspase-9活性均高于空白对照组(均P<0.05),而细胞荧光强度均高于空白对照组(均P<0.05);细胞凋亡率、胞浆AIF和细胞色素C含量、细胞Caspase-3和Caspase-9活性与黄芩素作用浓度均呈正相关(均P<0.05),而细胞荧光强度与黄芩素作用浓度呈负相关(P<0.05)。结论黄芩素对人表皮鳞癌A431细胞有显著的杀伤作用,激活线粒体凋亡通路可能是其发挥抑制皮肤鳞癌细胞作用的重要机制。Objective To investigate the effect of baicalein on mitochondrial apoptosis pathway in human epidermal squamous cell cancer A431 cells. Methods Human epidermal squamous cell cancer A431 cells were divided into blank control group and six groups of baicalein treatment,the baicalein treatment groups were given intervention with the corresponding concentrations of baicalein( 10,20,30,40,50 and 60 μg/ml,respectively),and the blank control group was given the equivalent volume of dimethyl sulphoxide( DMSO). The cell inhibition rate was detected in each group after 24,48 and 72 hours of intervention. Then another A431 cells were divided into blank control group and three groups of baicalein treatment,the baicalein treatment groups were given intervention with the corresponding concentrations of baicalein( 10,20 and 30 μg/ml,respectively),and the blank control group was given the equivalent volume of DMSO. After 24 hours of intervention,the apoptosis and mitochondrial membrane potential were detected,and the levels of cytoplasmic cytochrome C and apoptosis-inducing factor( AIF) as well as the activities of cysteinyl aspartate-specific proteinase( Caspase)-9 and Caspase-3 were determined. Results After24,48 and 72 hours of intervention,the half maximal inhibitory concentrations( IC50) of baicalein on A431 cells were 82.29 μg/ml,24.67 μg/ml and 18. 66 μg/ml,respectively; the total apoptosis rate,the contents of cytoplasmic AIF and cytochrome C,and the activities of Caspase-9 and Caspase-3 in the 10 μg/ml,20 μg/ml and 30 μg/ml baicalein treatment groups were all higher than those in the blank control group( all P〈0. 05),and the fluorescence intensity was higher in the baicalein treatment groups compared to the blank control groups( all P〈0. 05). The apoptosis rate,the contents of cytoplasmic AIF and cytochrome C,and the activities of Caspase-9 and Caspase-3 positively correlated with the inhibitory concentration of baicalein( P〈0. 05),while the fluorescence intensity negati
