恩度联合紫杉醇对三阴型乳腺癌移植瘤的影响

摘　　要：目的:通过观察人三阴型乳腺癌在裸鼠模型中血管正常化后对紫杉醇的反应,探讨其疗效是否优于紫杉醇.方法:将人三阴型乳腺癌MDA-MB-231细胞株种植于BALB/c-nu雌裸鼠右下腹部皮下,随机将符合入组标准的28只移植瘤荷鼠分成4组,每组7只,A组,每天每次腹腔内注射生理盐水(质量分数10 mg/kg),连续17 d;B组,每天每次腹腔注射恩度(质量分数10 mg/kg),连续17 d;C组,第6、12天腹腔注射紫杉醇(质量分数10 mg/kg),余同A组;D组,第6、12天腹腔注射紫杉醇(质量分数10 mg/kg),余同B组.测量移植瘤的长径和短径,进行病理学检测.结果:D组移植瘤体积小于A组、B组(P<0.05),C组移植瘤体积小于A组及B组(P<0.05),D组和C组移植瘤体积无差别(P=0.436).B组、C组、D组的抑瘤率分别是14.61%、39.08%、54.79%.解剖各组荷鼠未见远处转移病灶.病理检查微血管密度,D组<C组=B组<A组.Ki67,D组与C组<A组与B组,D组与C组无差别(P=0.649).结论:使用恩度后在时间窗内联合紫杉醇有明显抑瘤作用,但疗效未优于紫杉醇.Aim: To explore whether the efficacy of paclitaxel treatment after vascular normalization is better than paclitaxel alone by observing the response of human triple negative breast cancer( TNBC) to paclitaxel in nude mice. Methods: The human TNBC MDA-MB-231 cell line was transplanted into the right lower abdomen region of BALB/c-nu female nude mice. A total of 28 tumortransplanted mice,meeting the inclusion criteria,were randomly divided into 4 groups with 7 mice in each group. In group A,10 mg/kg/day saline was given for 17 consecutive days. In group B,the mice were given endostatin 10 mg/kg/day for 17 consecutive days. In group C,10 mg/kg/day saline was given for 17 consecutive days,and 10 mg/kg/day paclitaxel was given on the 6^(th) and 12^(th)days. In group D,the micewere given endostatin 10 mg/kg/day perfor 17 consecutive days,and 10 mg/kg/day paclitaxel was given on the 6^(th) and 12^(th)days. All the above-mentioned administration methods were intraperitoneal injections.The long and short diameters of all the transplanted tumors were measured on the 17 thday. Pathological and immunohistochemical examinations were employed. Results: The volume of transplanted tumor in group D was smaller than that in group A and group B( P < 0. 005),the volume of transplanted tumor in group C was smaller than that in group A and group B( P = 0. 05). There was no statistical difference between group D and group C( P = 0. 436). The tumor inhibition rates in group B,C and D were-14. 61%,39. 408%,54. 79%,respectively. There were nodistant metastasis or lesions found in all mice. MVD was: group D < group C = group B < group A. Ki67 was group D and group C < group B and group A,and there was no difference between group D and group C( P = 0. 649). Conclusion:Endostatin combined with paclitaxel chemotherapy in the normalized time window could effectively inhibit human triple negative breast cancer transplanted tumor,but the present study did not demonstrate that the efficacy is better than the use of paclitaxel alone.
