摘 要:目的为深入研究miR-30b-5p在乳腺癌形成和发展中的作用提供理论依据。方法原位杂交法(ISH)检测乳腺癌与癌旁组织中miR-30b-5p的表达。探针荧光定量PCR比较乳腺癌细胞系miR-30b-5p表达情况。软件预测miR-30b-5p的靶基因,进行KEGG及GO富集分析。结果 ISH显示乳腺癌组织中miR-30b-5p表达明显下调,与癌旁组织相比差异有统计学意义(P<0.01);miR-30b-5p的表达水平与TNM分期及远处转移等有关(P<0.01),而与年龄和淋巴结转移无关(P>0.05)。比较MCF-7,MDA-MB-231,MDA-MB-415,MDA-MB-453,和MDA-MB-468这5株乳腺癌细胞系miR-30b-5p表达情况,发现较正常乳腺组织呈不同程度的低表达,并且以侵袭性最强的MDA-MB-231细胞的表达最低(P<0.01)。在乳腺癌中,KEGG分析显示miR-30b-5p的靶基因参与了Pathways in cancer、P53signaling pathway、Wnt signaling pathway、Cell cycle等通路,GO分析显示miR-30b-5p具有细胞代谢调控、基因表达调控、RNA代谢调控等生物功能。结论 miR-30b-5p在乳腺癌中表达明显下调,可能通过参与Pathways in cancer、P53signaling pathway、Wnt signaling pathway、Cell cycle等通路,对乳腺癌细胞的代谢、基因表达等功能具有重要影响。Objective To provide theoretical guidance for further research on the role of miR-30 b-5 p in formation and development of breast cancer.Methods The miR-30 b-5 p in tissue chip by ISH was detected.The expression of miR-30 b-5 p in breast cancer cell lines was compared by fluorescence quantitative PCR.The target genes of miR-30 b-5 p were forecast.KEGG and GO enrichment analyses were carried on.Results MiR-30 b-5 p was down-regulated in breast cancer tissues,with statistical difference compared to that in adjacent tissues(P〈0.01);the expression levels of miR-30 b-5 p were found to be significantly associated with tumor node metastasis(TNM)staging and distant metastasis(P〈0.01),without association with age and lymphatic metastasis.The comparison of expressions of miR-30 b-5 p in MCF-7,MDA-MB-231,MDA-MB-415,MDA-MB-453 and MDAMB-468 breast cancer cell lines showed that expressions in comparative normal breast tissues were low and the MDA-MB-231 cells had the lowest expression(P〈0.01).The analysis of KEGG and GO indicated that miR-30 b-5 p was involved in pathways in cancer,P53 signaling pathway,Wnt signaling pathway and cell cycle and played a role in cell metabolism regulation,gene expression regulation,RNA metabolism and cellular biological function.Conclusion The expression of miR-30 b-5 p is down-regulated in breast cancer.It may play an important role in breast cancer cell metabolism,gene expression and so on through pathways in cancer,P53 signaling pathway,Wnt signaling pathway and Cell cycle.
