摘  要:目的探讨结肠癌组织中mi R-98的表达及作用机制。方法搜集40例结肠癌患者的肿瘤组织及癌旁组织标本,实时PCR检测mi R-98在组织中的表达。在HCT116细胞中分别过表达/沉默mi R-98后,MTT实验观察mi R-98对其增殖的影响;流式细胞技术分析mi R-98对细胞周期的影响;Hochest 33258染色判断mi R-98对其凋亡的影响。结果实时PCR检测发现,与癌旁组织比较,mi R-98在肿瘤中低表达(P=0.022);生存分析发现mi R-98表达低的患者5年生存率较低(P<0.05)。MTT检测发现mi R-98可以抑制细胞增殖。mi R-98过表达可以抑制细胞周期进程,促进HCT116细胞凋亡。结论 mi R-98可抑制结肠癌细胞增殖、促进细胞凋亡。mi R-98表达与结肠癌患者的生存期密切相关。Objective To investigate the expression and action mechanism of miR-98 in colon cancer tissues. Methods Tumor tissues and adjacent tissues were collected from 40 patients with colorectal cancer. The expression of miR-98 in tumor tissues and adjacent tissues was detected by real-time PCR. miR-98 was overexpressed or silenced in cells, and the effects on proliferation, cell cycle, and apoptosis were analyzed using MTF, flow cytometry, and Hoechst 33258 assays. Results Real-time PCR showed that the expression of miR-98 in tumor tissues was lower than that in adjacent tissues ( P = 0.022). The survival rate of patients with lower miR-98 expression was short- er than that of patients with higher miR-98 expression. The MTF assay showed that miR-98 overexpression inhibited the proliferation of HCT116 ceils. Hoechst 33258 staining showed that the overexpression of miR-98 could inhibit the cell cycle and promote the apoptosis of HCT116 ceils. Conclusion miR-98 can inhibit the proliferation and promote the apoptosis of colon cancer cells. The expression of miR- 98 is closely related to the survival of patients with colon cancer.
