摘 要:目的探讨微小RNA-1(miR-1)在心肺复苏术(CPR)后心搏骤停(CA)大鼠血浆中的表达及意义。方法32只SD大鼠随机均分为两组:CPR组采用窒息法建立CA大鼠CPR模型;对照组实施假手术。对照组于气管切开后及CPR组于自主循环恢复后24h取静脉血及心肌组织。采用qRT-PCR检测两组大鼠血浆miR-1的表达。测定两组大鼠血浆肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)和肌钙蛋白I(cTnI)含量,分析其与血浆miR-1表达的相关性。采用TUNEL法检测两组大鼠心尖部心肌细胞凋亡指数(AI),免疫组化染色法检测心肌Bcl-2蛋白表达。结果CPR组大鼠血浆miR-1、CK、CK-MB、cTnI和Bcl-2蛋白表达及AI均高于对照组(P<0.05)。CPR组大鼠血浆miR-1的表达量与CK、CK-MB、cTnI含量和AI呈正相关性(r=0.721、0.664、0.911和0.783,P<0.01),与Bcl-2蛋白表达呈负相关(r=-0.876,P<0.01)。结论miR-1可能参与了CPR后心功能障碍的发生,可能通过调控靶基因的表达促进心肌细胞凋亡。Objective To investigate the expression and significance of microRNA-1(miR-1)in plasma of the rats with cardiac arrest after cardiopulmonary resuscitation(CPR).Methods Thirty-two SD rats were equally randomized into two groups.The rats in CPR group were experienced cardiac arrest and CPR and those in control group were sham operated as model controls.The venous blood and myocardial tissues were collected from the rats in CPR group at 24 hours after spontaneous circulation recovery and those in control group after tracheotomy.Plasma level of miR-1 expression in both groups was detected by qRT-PCR.The concentrations of creatine kinase(CK),creatine kinase isoenzyme(CK-MB)and troponin I(cTnI)in two groups were measured as well.The correlation between miR-1 and these indexes was analyzed.The apoptotic index(AI)of myocardial apex was detected by TUNEL method.The expression of Bcl-2 protein was detected by immunohistochemical staining.Results The expressions of miR-1,CK,CK-MB,cTnI and Bcl-2 protein in CPR group were higher than those in control group(P〈0.05).Expression of miR-1 in CPR group was positively correlated with that of CK,CK-MB,cTnI and AI(r=0.721,0.664,0.911 and 0.783,P〈0.01),which was negatively correlated with Bcl-2 protein expression(r=-0.876,P〈0.01).Conclusion miR-1 may be involved in the occurrence of cardiac dysfunction after CPR,which may be associated with the promotion of cardiomyocyte apoptosis by regulating the expression of target genes.
