摘 要:目的:观察血红素加氧酶(heme oxyenase,HO)2种同工酶HO-1和HO-2在慢性脑血流灌注不足大鼠的肝脏中的动态变化并探讨其意义。方法:42只成年雄性SD大鼠,4月龄,体质量250~300 g,随机分为模型组和假手术组。假手术组只分离出双侧颈总动脉但不结扎,模型组行永久性双侧颈总动脉结扎术,然后将大鼠分别在术后1、7、14、21、28 d处死,分别用逆转录-聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)和Western blot检测HO-1和HO-2的表达,并检测HO的活性变化。结果:RT-PCR和Western blot显示,与假手术组比较,HO-1 mRNA和蛋白水平在术后1 d即有增加,并在第7天达到高峰(P=0.000,P=0.000);伴随着脑血流供应的代偿性恢复,HO-1 mRNA和蛋白的表达在术后14、21、28 d都逐渐降低,但是和假手术组相比,在术后14 d和21 d时,差异都有统计学意义(P=0.000,P=0.000)。这样的变化与HO的活性变化一致(P=0.000)。然而RT-PCR和Western blot发现HO-2 mRNA和蛋白水平在慢性脑血流灌注不足的肝脏中没有变化(P=0.604,P=0.329)。结论:血红素加氧酶和HO-1 mRNA以及蛋白的表达在慢性脑血流灌注不足的肝脏中增加,而HO-2 mRNA和蛋白的表达变化无统计学意义。提示HO-1通过增加血红素加氧酶活性保护肝脏免收慢性脑血流关注不足引起的损伤。Objective:To observe the dynamic change of heme oxygenase-1(HO-1)and heme oxygenase-2(HO-2)in the livers of the rats with cerebral hypoperfusion,and to explore their significance.Methods:Sixty 4-month-aged male SD rats,weighing 250 to300 g,were enrolled and divided into model groups and sham group.Model groups were performed by permanent bilateral common carotid arteries occlusion(2 vessel occlusion,2 VO)and randomly divided into 5 groups(1,7,14,21,28 d after 2 VO).The expressions of HO-1 and HO-2 at mRNA and protein levels were detected by RT-PCR and Western blot.HO activities were evaluated by ultraviolet specrophotometer.Results:RT-PCR and Western blot results showed that the expression of HO-1 mRNA and protein were increased in the livers on 1 d after 2 VO,and reached a peak on 7 d after 2 VO(P=0.000,P=0.000),accompanied by enhancement of HO activity(P=0.000).Nevertheless,RT-PCR and Western blot showed that the expression of HO-2 didn't change with time gradient(P=0.604,P=0.329).Conclusion:The activity of heme oxygenase and HO-1 mRNA and protein are increased in the livers of the rats with chronic cerebral hypoperfusion,but the expression of HO-2 at mRNA and protein level is not statistically changed,suggesting that HO-1 may play a protective role against liver injury induced by chronic cerebral hypoperfusion.
