摘  要:目的:探索由穿膜肽(transcriptional activator protein,TATp)和基质细胞衍生因子-1(stromal cell-derived factor-1,SDF-1)修饰的载多西紫杉醇超声造影剂对舌癌颈部淋巴组织转移灶的治疗效果。方法:以兔VX2舌癌颈淋巴转移模型、SCC-15细胞、超声造影剂为研究对象,将兔模型分为A、B、C、D组,分别用空白微泡(A)、靶向载药微泡(B)、单纯药物(C)和空白(D)处理;将细胞分为A、B、C、D、E、F组,分别用空白(A)、单纯药物组(B)、载药微泡(C)、靶向载药微泡(D)、靶向载药微泡+超声(E)和超声(F)处理细胞,通过测量淋巴结体积、免疫组化和细胞凋亡检验实验结果。结果:超声检查见颈部转移性淋巴结,实验中各组的肿瘤抑制率分别为5.3%、25.4%、22.3%、0.0%,组间比较:PAD=0.436,PBD=0.010,PCD=0.008,PBC=0.723;各组p53阳性率分别为(29.8±3.3)%,(3.8±2.8)%,(3.8±3.1)%,(30.8±3.6)%,组间比较:P_(AD)=0.663,P_(BD)=0.000,P_(CD)=0.000,P_(BC)=1.000。细胞实验中各组细胞凋亡率分别为(17.82±1.39)%,(57.61±2.37)%,(31.87±2.18)%,(46.28±1.65)%,(67.34±2.22)%,(14.17±8.13)%,组间比较P_(AB)=0.000,P_(AC)=0.001,P_(AD)=0.000,P_(AE)=0.000,P_(AF)=0.486,P_(EB)=0.007,P_(EC)=0.000,P_(ED)=0.000,P_(EF)=0.000。结论:TATp和SDF-1修饰的载药超声微泡在治疗中有一定作用,能为舌癌颈淋巴结转移灶的临床治疗提供参考。Objective:To explore the imaging effect and treatment effect of the ultrasonic microbubble modified by transcriptional activator protein(TATp)and stromal cell-derived factor-1(SDF-1)carrying docetaxel lipid in metastatic lymph node lymph node of tongue cancer.Methods:A control study was made with the VX2 cervical lymph node metastasis in rabbits model,SCC-15 cell and the ultrasonic microbubble modified by TATp and SDF-1 carrying docetaxel.The rabbit model was divided into A,B,C,D groups,respectively dealing with blank microbubbles(A),targeted microbubbles(B),drug(C)and blank(D).The cells were divided into:A,B,C,D,E,F groups,respectively dealing with blank(A),drug group(B)and microbubbles(C),targeted microbubbles(D),targeted microbubbles + ultrasound(E)and ultrasound(F).The results were tested by measuring lymph node's volume,immunohistochemistry,and apoptosis.Results:The ultrasonography revealed metastatic cervical lymph nodes.The tumor inhibitory rates in each group were 5.3%,25.4%,22.3%,0.0%.Comparison between groups showed that:PAD=0.436,PBD=0.010,PCD=0.008,PBC=0.723.The positive rates of p53 were(29.8±3.3)%,(3.8±2.8)%,(3.8±3.1)%,(30.8±3.6)%.Comparison between groups were PAD=0.663,PBD=0.000,PCD=0.000,PBC=1.000.The apoptotic rates were(17.82±1.39)%,(57.61±2.37)%,(31.87 ±2.18)%,(46.28 ±1.65)%,(67.34±2.22)%,(14.17±8.13)%.Comparisons between groups were PAB=0.000,PAC=0.001,PAD=0.000,PAE=0.000,PAF=0.486,PEB=0.007,PEC=0.000,PED=0.000,P_(EF)=0.000.Conclusion:Ultrasounic microbubble modified by TATp and SDF-1 play a certain role in the treatment,and can provide reference for the clinical treatment of cervical lymph node metastasis.
