摘　　要：目的观察脂肪干细胞对肿瘤坏死因子-α(TNF-α)诱导的牙周膜干细胞炎症因子表达的影响,以期为脂肪干细胞应用于牙周炎治疗提供理论指导。方法培养、鉴定脂肪干细胞和牙周膜干细胞。采用Transwell细胞共培养系统,上、下室均接种第3代细胞。实验分3组:A组:牙周膜干细胞对照组,上、下室均接种牙周膜干细胞;B组:肿瘤坏死因子-α诱导组,上、下室均接种牙周膜干细胞,下室加入10ng/m L肿瘤坏死因子-α;C组:肿瘤坏死因子-α联合脂肪干细胞干预组,上室脂肪干细胞,下室牙周膜干细胞,下室加入10ng/m L肿瘤坏死因子-α。逆转录聚合酶链反应检测牙周膜干细胞白介素-6、白介素-8和肿瘤坏死因子-α的mRNA表达。结果 B组较A组白介素-6、白介素-8和肿瘤坏死因子-α的表达均增加,且均在2h时达到峰值(P〈0.05)。2h时C组较B组白介素-6、白介素-8和肿瘤坏死因子-α的表达明显下降(P〈0.05)。结论脂肪干细胞能抑制TNF-α诱导的牙周膜干细胞的炎症反应。Objective To investigate the effect of adipose derived stem cells （ADSCs）on inflammatory factors expression of periodontal ligament stem ceils （PDLSCs） induced by tumor necrosis factor. Methods PDLSCs and ADSCs were co-cultured and identified by transwell assay. The cells were divided into three groups, PDLSCs control group（ group A ）, TNF-α stimulation group （ group B ）, TNF-α plus ADSCs group （ group C ）. The expression levels of interleukin-6 （IL-6）, interleukin-8 （IL-8）, tumor necrosis factor （TNF-α） mRNA of PDLSCs were detected by RT-PCR. Results Compared with group A, the expression of IL-6, IL-8 and TNF-α in group B were increased and reached the peak at 2h （ P 〈 0.05 ）. Compared with group B, the expression of IL-6, IL-8 and TNF-α was decreased in group C at 2h（ P 〈 0.05 ）. Conclusion ADSCs could inhibit the inflammatory response of PDLSCs induced by tumor necrosis factor.
