摘  要:目的研究氢气水(H_2水)滴眼对大鼠碱烧伤诱导角膜新生血管(corneal neovascularization,CNV)的抑制作用并探讨H2水抑制CNV的机制。方法 SPF级SD大鼠48只48眼,成功建立碱烧伤诱导CNV模型,随机分为空白组、对照组和治疗组。治疗组给予1.6×10^(-6)H_2水滴眼,对照组给予妥布霉素地塞米松眼液滴眼,空白组不做任何处理。分别在模型建立后第1天、第3天、第7天、第14天测量CNV长度,计算CNV面积。碱烧伤后第14天深麻醉下取大鼠角膜及房水,免疫组织化学染色法检测角膜组织中PEDF蛋白表达,RT-PCR法检测角膜组织中PEDF mRNA表达,ELISA检测房水内TNF-α蛋白表达。结果角膜碱烧伤后第3天、第7天、第14天,治疗组新生血管面积分别为(3.26±1.82)mm^2、(8.59±3.98)mm^2和(3.24±2.34)mm^2;对照组分别为(4.27±2.68)mm^2、(6.44±4.67)mm^2和(15.98±4.75)mm^2;空白组分别为(9.49±2.79)mm^2、(17.71±4.06)mm^2和(25.35±1.40)mm^2。碱烧伤后第3天、7天和14天,治疗组和对照组均小于空白组,差异均有显著统计学意义(均为P<0.01);碱烧伤后第3天和第7天,治疗组与对照组差异均无统计学意义(均为P>0.05);碱烧伤后第14天治疗组明显小于对照组,差异具有显著统计学意义(P<0.01)。碱烧伤后第14天,免疫组织化学染色结果显示,治疗组角膜上皮层PEDF蛋白表达较对照组和空白组明显增强,基质层新生血管极少;RT-PCR检测结果显示,治疗组PEDF mRNA表达明显高于对照组和空白组,差异均有显著统计学意义(均为P<0.01);ELISA检测结果显示,治疗组房水中TNF-α蛋白含量明显低于对照组与空白组,差异均有显著统计学意义(均为P<0.01)。结论 1.6×10^(-6)H_2水滴眼能有效抑制大鼠碱烧伤诱导的CNV。Objective To observe the inhibitory effects of hydrogen on corneal( C NV) induced by alkali burn in rats and its underlying mechanisms Methods A total of 48 SPF-grade SD rats( 48 eyes) w ere selected and randomly divided into blank group,control group and treatment group. T hen the model of C NV w as established successfully induced by alkali burn in these animals. T he treatment group w as given 1. 6 × 10^(-6) hydrogen eye drops,the control group received tobramycin dexamethasone eye drops,and blank group left untreated. And C NV length w as measured on day1,3,7 and 14 after model establishment,follow ed by calculation of the C NV area. T he corneal and aqueous humor of the rats w as removed on day 14 after alkali burn. T hen PEDF protein w as detected by immunohistochemical staining,and RT-PC R w as used to detect the expression of PEDF mRNA in the cornea. Additionally,T NF-α in the aqueous humor w as detected by enzyme e-linked immunosorbent assay( ELISA). Results T he area of corneal neovascularization on day 3,7 and 14 after alkali burn w as( 3. 26 ± 1.82) mm^2,( 8. 59 ± 3. 98) mm2 and( 3. 24 ± 2. 34) mm^2 in the treatment group,r espectively,( 4. 27 ± 2. 68) mm^2,( 6. 44 ± 4. 67) mm2 and( 15. 98 ± 4. 75) mm^2 in the control group,r espectively,( 9. 49 ± 2. 79) mm^2,( 17. 71 ± 4. 06) mm2 and( 25. 35 ± 1. 40)mm^2 in blank group,r espectively. T he C NV areas of the treatment group and the control group w ere less than that of the blank group on day 3,7 and 14 after alkali burn,and the differences w ere statistically significant( all P < 0. 01),but there w as no significant difference between the treatment group and the control group on day 3,7 after alkali burn( P > 0.05),while the area of the treatment group was significantly less than that of the control group on 14 th day,and the difference was statistically significant( P < 0. 01).
