摘  要:目的探究结肠癌远处转移小鼠模型血清miR-449的表达及意义。方法皮下注射结肠癌HCT116细胞建立小鼠结肠癌远处转移模型,实时定量PCR法检测转移组、未转移组和正常组小鼠血清miR-449的表达情况。绘制ROC曲线,观察miR-449评判结肠癌转移的灵敏度与特异度。结果相比于正常小鼠,未转移小鼠中miR-449的表达水平并未明显增高(P>0.05);转移小鼠组小鼠的miR-449的表达水平较未转移组升高20.3倍,差异有显著显著性(P<0.01)。ROC曲线显示,血清miR-44表达水平评判结肠癌转移的灵敏度91%,特异度为94%。结论血清miR-449表达水平可为临床判定结肠癌远处转移提供新的实验线索,为临床深入研究结肠癌的诊断和治疗提供新的实验依据。Objective To explore the experimental significance of expression level of serum miR-449 in mouse models with distant metastasis of colon cancer.Methods The mouse models with distant metastasis of colon cancer were established and the expression of serum miR-449 in different groups(metastasis group,non-metastasis group and control group) of mice had been detected by quantitative PCR.Results In comparison with normal mice,mice in non-metastasis group did not showed elevated expression level of miR-449(P > 0.05).The level of miR-449 expression in mice of metastasis group had been increased 20.3 times than that in mice of non-metastasis group,and the difference was significant(P < 0.01).ROC curve showed that the sensitivity and specificity of expression level of serum miR-449 in judgement of metastasis of colon cancer were 91% and 94% respectively.Conclusion The expression level of serum miR-449 l can provide new experimental clues for clinical judgement of distant metastases of colon cancer,and it provides new experimental basis for clinical diagnosis and treatment of patients with colon cancer.
