摘 要:目的探讨自噬标记蛋白Beclin1在宫颈癌组织中的表达检测及其与肿瘤增殖侵袭活性的关系。方法回顾性收集2014年1月至2016年12月间接受手术治疗的原发性宫颈癌患者80例,采用免疫组织化学法检测宫颈癌组织、癌旁组织中Beclin1蛋白表达情况,根据宫颈癌组织中Beclin1蛋白表达进一步将其分为Beclin1蛋白阳性组、Beclin1蛋白阴性组。采用荧光定量PCR法测定宫颈癌组织中增殖、侵袭基因表达量。结果宫颈癌组织中Beclin1蛋白阳性表达率显著低于癌旁组织(P<0.05)。Beclin1蛋白阴性组宫颈癌组织中增殖基因GBP1 mRNA的表达量低于Beclin1蛋白阳性组,STAT3、RPL6、YAP1、EZH2 mRNA的表达量高于Beclin1蛋白阳性组;侵袭基因CXCR7、Sox2、Survivin、Rab11、Rb Ap48 mRNA的表达量高于Beclin1蛋白阳性组,p27 mRNA表达量低于Beclin1蛋白阳性组(P<0.05)。结论宫颈癌组织中自噬标记蛋白Beclin1阳性表达减少,可直接导致肿瘤细胞增殖侵袭活性增高。Objective To study the expression of autophagy marker protein Beclin1 in cervical carcinoma and its correlation with tumor proliferation and invasion.Methods 80 cases of cervical carcinoma patients in our hospital during January 2014 to December 2016,the expression of Beclin1 protein in cervical cancer tissue and adjacent normal tissue was detected by immunohistochemistry,according to Beclin1 expression in cervical cancer tissue all patients were further divided into Beclin1 positive group and Beclin1 negative expression group.The expression of proliferating and invasive genes in cervical cancer tissues were determined by fluorescent quantitative PCR.Results The positive rate of Beclin1 protein in cervical cancer tissues was lower than that in adjacent tissues(P < 0.05).The expression of GBP1 gene in Beclin1 protein negative group was lower than that in Beclin1 positive expression group,STAT3,RPL6,YAP1,EZH2 mRNA were higher than those in Beclin1 positive expression group;expression of invasion genes such as CXCR7,Sox2,Survivin,Rab11,Rb Ap48 mRNA were higher than those in Beclin1 protein positive group(P < 0.05).Conclusion The positive expression of autophagy marker protein Beclin1 in cervical cancer tissue is decreased,which can directly lead to the proliferation and invasion of tumor cells.
