摘 要:目的:基于治疗前、后IBS大鼠pPKCγ和ERK1基因在背根神经节的表达,探讨安肠汤治疗肝郁脾虚型IBS的机制。方法:40只清洁雄性SD大鼠随机分为4组,10只/组,正常组、模型组、西药组、中药组。采用番泻叶+束缚应激法造模。造模后治疗组和对照组分别予安肠汤煎剂及得舒特片2周,用腹部回缩反射评估大鼠内脏敏感性,用PCR和Western-blot技术检测pPKCγ和ERK1基因与蛋白表达。结果:模型组背根神经节中的pPKCγ和ERK1基因表达与另外3组对比明显增高,差异有统计学意义,P〈0.05;正常组与西药组及中药组比较,P〈0.05,有统计学意义;西药组和中药组相比,P〉0.05,无统计学意义。结论:说明pPKCγ和ERK1的基因参与肝郁脾虚型IBS大鼠疼痛信号外周敏化机制;中药安肠汤可以有效调控pPKCγ和ERK1基因在背根神经节的表达,从而达到缓解腹痛的临床效应机制。Objective: Based on the IBS rat pPKCγ and ERK1 gene expressions respectively in dorsal root ganglia before and after treatment to explore the mechanism of Anchang Decoction in treatment of liver depression and spleen deficiency type of IBS. Methods: Forty clean male SD rats were randomly divided into 4 groups,normal group,model group,western medicine group,TCM group,10 rats/group. The model of restraint stress + Fangxieye method. The TCM group and the western medicine group were given Anchang Decoction and Dictel for 2 weeks. Results: The expressions of pPKCγ and ERK1 in the dorsal root ganglia of the model group were significantly higher than those in the other three groups,P 〈 0. 05. The western medicine group and TCM group had no significance,P 〉 0. 05. Conclusion: The pPKCγof ERK1 and gene were involved in the liver stagnation and spleen deficiency type IBS rat pain signals from the central sensitization mechanism. Anchang Decoction can effectively regulate pPKCγ and ERK1 gene in dorsal root ganglia expression so as to alleviate the pain of clinical effect mechanism.
