摘  要:细胞代谢在癌症起始和转移性进展疾病中起到核心作用,代谢转换为有氧糖酵解和乳酸产生增加是侵袭性肿瘤细胞的特征,并且是肿瘤反应和治疗结果的共同决定因素。由于细胞代谢是相互依赖的途径的复杂网络,局部改变将对整体肿瘤代谢产生影响,而肺癌的发生和发展是一个多步骤过程,其特征在于许多遗传和表观遗传改变的积累,包括致癌基因和肿瘤抑制基因的改变,导致关键细胞-调节和生长-控制通路的改变。对肺癌相关癌基因和肿瘤抑制基因与中心碳代谢的研究做一论述,以期在葡萄糖转运和能量代谢途径中,挖掘可以阻断失调的癌基因和/或恢复功能丧失的肿瘤抑制基因的途径,以探索肺癌治疗的新方法,并作为开发新的抗肺癌转移疗法有价值的靶标。Cell metabolism plays a central role in cancer initiation and metastatic progressive disease. The metabolism is converted to aerobic glycolysis and increased lactate production,which is the characteristic of invasive tumor cells,and common determinants of tumor response and treatment outcome. Since cell metabolism is a complex network of interdependent pathways,local changes will have an impact on overall tumor metabolism,however,the occurrence and development of lung cancer is a multi-step process,characterized by the accumulation of a number of genetic and epigenetic changes,including the changes of oncogenes and tumor suppressor genes,leading to changes in key cellular-regulatory and growth-control pathways. In this article,we summarized the lung cancer related oncogenes and tumor suppressor genes as well as central carbon metabolism,digging a pathway that can block the disordered oncogene and/or restore the tumor suppressor gene that has lost function in order to explore new approaches to the treatment of lung cancer,and as a valuable target in the development of new anti-lung cancer therapies.
