摘　　要：目的:阐述印迹基因PEG3在肾细胞癌中的表达及意义。方法:利用Oncomine数据库及GEPIA分析PEG3在肾透明细胞癌组织中的表达情况;利用KM-Plotter做PEG3与肾透明细胞癌患者生存期的相关性分析,经Meth HC分析PEG3启动子区甲基化水平,String-DB数据库分析PEG3相互作用蛋白;通过The Human Protein Atlas分析PEG3蛋白在肾透明细胞癌中的蛋白表达情况。结果 :在mRNA水平上,PEG3在肾透明细胞癌中较正常肾组织显著低表达,表达水平与肾癌预后呈负相关(Log-rank P=0.001 1),PEG3蛋白在肾透明细胞癌中也较正常肾脏组织低表达,且其启动子甲基化水平则显著增高。与PEG3相关蛋白有NLRP7、MEST、SIAH1、PEG10、CDC25 B等,可能参与细胞有丝分裂周期调节及遗传印迹等细胞功能。结论 :通过肿瘤基因数据库信息挖掘发现,PEG3在肾透明细胞癌组织中呈低表达,且与患者生存期有关,将为后续的肿瘤研究提供重要的理论依据。Objective：To explore the expression and significance of imprinted gene PEG3 in clear cell renal cell carcinoma.Methods：The expression of PEG3 in clear cell renal cell carcinoma was mined in Oncomine database and GEPIA.The correlation between PEG3 and the survival time of patients with clear cell renal cell carcinoma was analyzed by KM-Plotter.The methylation level of PEG3 promoter region was used by MethHC,and the PEG3 - interacting proteins were analyzed by String-DB database.The expression of PEG3 protein in normal renal tissue and clear cell renal cell carcinoma was detected by Human Protein Atlas.Results：At the mRNA level,PEG3 was lower in clear cell renal cell carcinoma than normal renal tissue,and the expression level was negatively correlated with the prognosis of renal cell carcinoma （Log-rank P=0.001 1）,the expression of PEG3 protein in clear cell renal cell carcinoma was also lower than that in normal renal tissue.The methylation of PEG3 promoter in clear cell renal cell carcinoma was increased.PEG3 related proteins are NLRP7,MEST,SIAH1,PEG10,CDC25B,etc,may be involved in cell mitotic regulation and genetic imprinting and other cellular functions.Conclusion：PEG3 was decreased in clear cell renal cell carcinoma and was related to the patients＇ survival,it would be an important theoretical support for the further research.
