摘  要:为比较填充不同填料(悬浮填料和弹性填料)生物反应器的抗冲击负荷能力,研究了不同水力停留时间(14.80 h、11.90 h、9.89 h和7.99 h)下两组反应器的处理性能及附着生物膜的特性,通过提取基因组DNA,采用PCR扩增与高通量测序技术对水力停留时间为7.99 h工况下两者菌群测序并进行了操作单元(OTU)聚类分析、多样性分析和分类学分析。结果表明,不同水力停留时间下弹性填料附着的生物膜量和胞外多聚物(EPS)质量比均高于悬浮填料,生物膜结构较为稳定;弹性填料和悬浮填料富集微生物菌群的高通量测序分析均得到优化序列23 129条,测序覆盖深度都在99%左右,经过97%相似度归并后分别得到375个OTUs和307个OTUs,与悬浮填料相比,弹性填料附着的生物膜内硝化菌和反硝化菌丰度均较高,这在一定程度上表明弹性填料在处理污染河水时具有较强的抗冲击负荷能力。This paper is to present the results of its investigation of the treated effects and the characteristic features of the biofilm fillings through a treatment of 4 kinds of different hydraulic retention time( 14. 80 h,11. 90 h,9. 89 h and 7. 99 h). At the same time,it has also made a comparison of the shock resistance capacity of the 2 kinds of materials used as the biofilm carriers respectively in lab-scale bioreactors. Besides,we have also adopted the PCR amplification and high-throughput sequencing from the extracted genomic DNA to disclose the similarities and differences of the biofilms attached on the suspended filler and elasticity plastic filler. At the same time,we have also conducted the hierarchical clustering of the operational units and the analysis of the diversity and taxonomy. All the results of the aforementioned experimental results show the start-up speed of the bioreactor with the plastic elasticity filler tends to be greater as compared with that of the bioreactor of the suspended filler,which is likely to shorten the start-up stage of its earlier maturation in removing NH_4~+-N and CODMn. And,then,in the stable-steady running stage,the removing efficiencies of NH_4~+-N and CODMnin the 2 bioreactors can both be elevated up to the rates of 97% and 80%,respectively. However,in general,the average removal efficiency of NH_4~+-N in the bioreactors with elasticity plastic filler and suspended filler may tend to drop from 98. 18%,98. 76% to78. 32%,87. 63%,whereas the average removal efficiency of CODMnmay tend to fall from 83. 69%,84. 52% to 60. 90%,64. 71% accordingly with the hydraulic retention time( HRT)cutting down from 14. 80 h to 7. 99 h,which indicates that the effluent quality of the bioreactor with elasticity plastic filler can be taken as being superior to that of the bioreactor with the suspended filler.
