摘 要:为探讨巨噬细胞加帽蛋白(Cap G)在结直肠癌组织中的表达及对癌细胞增殖、迁移的影响,选取2016年2月至2017年2月我院收集的91份结直肠癌标本,同时选取癌旁正常组织(>5 cm)作为对照,采用免疫组化法检测Cap G表达;同时选用结直肠癌HCT116细胞分为空白对照组、阴性对照组和观察组,阴性对照组转染阴性序列,观察组转染靶向Cap G序列,采用MTT法和Transwell细胞迁移实验检测细胞增殖及迁移能力。结直肠癌组织Cap G阳性表达率为76.92%,明显高于癌旁正常组织,差异比较有统计学意义(p<0.05);不同癌变部位、分化程度及TNM分期患者Cap G阳性表达率比较差异有统计学意义(p<0.05);空白对照组、阴性对照组和观察组转染后A值比较差异无统计学意义(p>0.05);观察组穿膜细胞数分别为(55.20±8.42)个,明显低于空白对照组和阴性对照组(p<0.05);空白对照组和阴性对照组穿膜细胞数比较差异无统计学意义(p>0.05)。结直肠癌组织中Cap G表达明显升高,表达水平与癌变部位、分化程度及TNM分期有关,同时Cap G表达与结直肠癌细胞迁移有关。To investigate the expression of macrophage capping protein(Cap G) in colorectal carcinoma and its effects on proliferation and migration of cancer cells, 91 specimens of colorectal cancer tissue were collected from February 2016 to February 2017 in our hospital, and the adjacent normal tissues(〉5 cm) were selected as control group and the expression of Cap G was detected by immunohistochemistry. Meanwhile, colorectal cancer HCT116 cells were divided into control group, negative control group and observation group. The negative control group was transfected with negative sequence, and the observation group was transfected into the target Cap G sequence.Cell proliferation and migration ability were detected by MTT method and Transwell cell migration method. The positive expression rate of Cap G in colorectal carcinoma was 76.92%, which was significantly higher than that in adjacent normal tissues, and the difference was statistically significant(p〈0.05). There was a statistically significant difference in the positive expression rate of Cap G between different cancer sites, differentiation and TNM stages(p 〈0.05). The difference of A value in control group, negative control group and the observation group after transfection was not statistically significant(p〉0.05). The number of transmembrane cells in observation group was(55.20±8.42), which was significantly lower than that in the control group and the negative control group(p〈0.05).However, there was no significant difference in number of transmembrane cells between the control group and the negative control group(p〈0.05). The expression of Cap G in colorectal cancer significantly increased, and the expression of Cap G was related to the location of the tumor, the degree of differentiation and TNM stage, and the expression of Cap G was related to the migration of colorectal cancer cells as well.
