摘  要:为从分子水平了解牛乳头瘤病毒(bovine papillomavirus,BPV)贵州镇宁GZ01流行株全基因组序列、结构特征及遗传变异情况,本研究选取贵州镇宁患病牛皮肤肿瘤样物,以乳头瘤病毒L1基因的简并引物MY11/MY09进行PCR扩增,测序后并确定其基因型。根据Gen Bank中BPV参考株设计6对特异性引物,经扩增、测序和拼接后获得BPV流行株全基因组序列。结果序列分析表明,GZ01流行株全基因组长为7 944 bp,具有BPV2基因型的结构特征,与Gen Bank收录的牛乳头状瘤病毒2型基因型参考株BPV2、BPV2-SW01和BPV2-AKS01的核苷酸同源性高达98.8%、99.7%和99.4%,同时与BPV13、BPV1和BPV14的基因型参考株进化关系也比较最近,同属Dalte属。因此,贵州镇宁GZ01株为BPV2基因型,GZ01株为贵州地区首次检测确认并测定了全基因组序列的牛乳头瘤病毒。本研究为贵州地区的牛乳头状瘤的病原鉴定、流行规律、遗传变异以及科学的防控提供了理论依据。This experiment aimed to study the complete genomic sequence, structural characteristics, and genetic variation of the bovine papillomavirus 2 genotype(BPV2) GZ01 strain at the molecular level, genotyping of this strain from the skin samples of cattle in Zhenning, Guizhou. The strain was detected by the polymerase chain reaction with MY11/MY09 primers. After sequencing, its genotype was determined. According to the design of6 pairs of specific primers in the BPV Gen Bank reference strain, the whole genome sequence of BPV gene was obtained by amplification, sequencing and splicing. The sequence analysis showed that the Guizhou Zhenning GZ01 strain was BPV2 genotype, and the length of gene group was 7 944 bp, with structural features of the BPV2 genotype. The nucleotide homology was 98.8%, 99.7% and 99.4% compared with reference strains BPV2,BPV2-SW01 and BPV2-AKS01,at the same time it had close evolutionary relationship with genotype BPV13,BPV1 and BPV14, and they all belonged to the genus Dalte. Therefore, Zhenning GZ01 strains in Guizhou were BPV2 genotype, GZ01 strains were detected for the first time in Guizhou to confirm and determine the whole genome sequence of bovine papilloma virus. This study provided the theoretical basis for pathogen identification,epidemic regularity, genetic variation and scientific prevention and control of bovine papilloma in Guizhou area.
