补肾益气明目方组方药物治疗视神经萎缩靶向机制初探

摘　　要：目的:探讨补肾益气明目方组方药物干预视神经萎缩(optic atrophy,OA)的靶向机制。方法:采用数据挖掘方法,利用药物靶点数据库和疾病靶标数据库分别筛选药物组分和靶点以及OA疾病靶标分子并进行文献回顾分析。结果:该经验方10味中药有效组分中的木犀草素、原儿茶酸、槲皮素、维生素A和芝麻脂素可直接扰动OA靶标分子,主要涉及的5个靶标分子分别为EGFR、KIF21A、GJA1、ACOX1、G6PD,呈现多有效组分多靶点干预OA特点。结论:补肾益气明目方组方药物可能通过木犀草素、原儿茶酸、槲皮素、维生素A和芝麻脂素等有效组分对OA进行直接干预,其药理机制可能涉及促神经细胞的生成及稳态、胞间信号传导及运输动力调控、抑制退行进程、抗氧化等神经再生和神经保护的多个机制。objectire To analyze targeted treatment on OA （optic atrophy） by Bu Shen Yi Qi Ming Mu formula. Methods We employed data mining analysis technology based on molecular mechanisms research strategy directed by biological activity. Drug targets of the formula components and disease targets of OA were extracted by using of related database. Common targets of both formula effective components and OA disease were reviewed by literature briefly. Results The five effective components of formula such as luteolin, protocatechuic acid, quercetin, vitamin A and sesamin were identified, which could disturb the targets of OA directly. Five target molecules of OA were locked; they were EGFR, KIF21A, GJA1, ACOX1 and G6PD. A typical feature that multi-components-targets of formula for treating OA was shown in details. Conclusion Direct interfere on OA by effective components of Bu Shen Yi Qi Ming Mu formula might mainly involve luteolin, protocatechuic acid, quercetin, vitamin A and sesamin. Pharmacological mechanisms were probably associated with neuroprotection and nerve regeneration deriving from promotion of nerve cells formation and homeostasis, regulation of intercellular signal transduction and transport power, suppression of the regression process and anti-oxidation.
