摘 要:目的研究地榆皂苷Ⅰ对人甲状腺乳头状癌BCPAP细胞作用及相关的分子机制。方法 MTT法检测地榆皂苷Ⅰ对BCPAP细胞增殖的作用;AnnexinⅤ/PI双染法检测细胞凋亡;Western blot法检测凋亡相关蛋白表达水平;JC-1染色检测线粒体膜电位;试剂盒检测Caspase活性。结果地榆皂苷Ⅰ对BCPAP细胞增殖有显著抑制作用,且呈浓度依赖性(P<0.05),IC_(50)为28.99μmol/L。地榆皂苷Ⅰ可显著诱导BCPAP细胞凋亡,且凋亡率随着药物浓度的升高而显著升高,呈剂量依赖性。地榆皂苷Ⅰ可提高Bax/Bcl-2比率,降低线粒体膜电位,促使细胞色素C从线粒体内的释放,提高Caspase-9和Caspase-3活性。结论地榆皂苷Ⅰ通过激活线粒体通路导致人甲状腺乳头状癌BCPAP细胞增殖受到抑制并诱导细胞凋亡。Objective To observe the effects and underlying molecular mechanisms of Ziyugly- coside [ (ZI ) on human thyroid papillary carcinoma BCPAP cells. Methods The proliferation inhibito- ry of ZI on BCPAP cells was detected by MTT assay. Cell apoptosis was detected by flow cytometry with double staining of Annexin V/PI. The expression levels of apoptosis-related proteins were performed by Western blot. Mitochondrial membrane potential (MMP) was detected by flow cytometry with JC-1 staining. Caspase activity was detected by reagent kit. Results ZI significantly inhibited the proliferation of BCPAP cells in a dose-dependent manner (P 〈0.05, IC50 value: 28.99 μmol/L). Z [ could positively induce BCPAP cell apoptosis in a dose-dependent manner. Z I could increase Bax/Bcl-2 ratio, decrease the level of MMP, promote the release of cytochrome C from mitochondria, and elevate activities of Caspase-3 and Caspase-9. Conclusion ZI could inhibit the proliferation of human thyroid papillary carcinoma BCPAP cells through activating mitochondria-dependent pathway and induce cell apoptosis.
