摘 要:采用单因素法优化了乙醇浸提法提取海带岩藻黄素的工艺条件,对提取物进行了红外光谱分析,并测定分析提取物的抗氧化活性及降血糖活性。研究结果表明,乙醇浸提法提取海带岩藻黄素的优化条件为:固液比1∶40,温度50℃,时间2 h,提取2次,提取率为0.222 mg/g,纯度为90.6%;红外光谱在3 388 cm-1处出现羟基的特征吸收峰,羰基的吸收峰在1 721 cm-1处;岩藻黄素对H2O2羟基自由基的清除率达81.6%,对α-葡萄糖苷酶的抑制率达64.7%。The single-factor experiment method was used to optimize the conditions of the ethanol extraction method in order to extract fucoxanthin from Laminaria japonica, and the fucoxanthin was analyzed by infra-red spectrum. The antioxidation of fucoxanthin was researched by observing its scavenging ability on hydroxyl radical of H2 O2, and its inhibition effect on the activity of α-glucosidase was also studied. The result of research shows that the optimal parameters with ethanol extraction method for extracting fucoxanthin from Laminaria japonica were as follows: the solid-liquid ratio was 1:40; the optimized temperature was at 50 ℃; extraction time was 2 h; extraction times of 2. Under these optimized process, the extraction rate was 0.222 mg/g and the purity was 90.6 %. The characteristic absorption peak of hydroxyl appeared at 3388 cm-1, and the characteristic absorption peak of carbonyl appeared at 1721 cm-1. The antioxidation experiment of the fucoxanthin showed that its scavenging rate on hydroxyl radical of H2 O2 could reach up to 81.6 %, and its inhibition rate on the activity of α-glucosidase could reach up to 64.7 %.
