摘 要:目的:本文研究了白背三七中多种具有降血糖作用的黄酮类药物对牛血清白蛋白(BSA)结构的影响,分析了其作用位点、结合常数等。方法:利用紫外光谱法、荧光光谱法等,在生理条件pH=7.4的条件下,研究了白背三七中木犀草素、槲皮素、山奈酚在常温(298K)下分别与牛血清白蛋白的相互作用。结果:木犀草素、槲皮素、山奈酚与牛血清白蛋白结合作用较强,在常温(298K)时,结合常数KA分别为1.16×10~6、7.64×10~5、1.32×10~6L/mol;它们都使BSA的构象发生了变化。结论求出了三种黄酮成分与BSA的结合常数及作用位点,证明黄酮与BSA的作用使BSA的构象发生变化。Purpose: In this paper, the efects of several favonoids on the structure of bovine serum albumin (BSA) were studied, and their action sites and binding constants were analyzed. Method: The interaction of luteolin, quercetin and kaempferol in white back radix notoginseng with bovine serum albumin at room temperature (298k) by ultraviolet spectroscopy and fuorescence spectroscopy at pH=7.4. Result: The interaction is strong between luteolin, quercetin, kaempferol and bovine serum albumin. At room temperature (298K), the binding constant KA were 1.16× 106, 7.64× 105 and 1.32× 106 l / mol. They all change the conformation of BSA. Conclusion: The binding constants and action sites of three favonoids and BSA were determined. And it was proved that the conformation of BSA was changed by the action of favonoids and BSA.
