摘  要:建立HPLC法测定呋喃苦参黄连素片中四种活性成分(呋喃唑酮、盐酸小檗碱、苦参碱和氧化苦参碱)含量的方法.呋喃唑酮与盐酸小檗碱含量测定采用氨基色谱柱:Inertsil NH_2 5μm 4.6×250 mm,流动相为乙腈,无水乙醇,2%磷酸溶液,呋喃唑酮和盐酸小檗碱检测波长分别为365 nm和350 nm.苦参碱及氧化苦参碱含量测定采用十八烷基硅烷键合硅胶柱:Promosil C_(18)5μm 4.6×250 mm;流动相为甲醇∶磷酸盐缓冲溶液(7∶93);检测波长为220 nm.流速为1.0 mL·min^(-1);柱温为30℃.实验结果表明分析时间短,色谱峰分离良好,分离度和重复性好,呋喃唑酮、盐酸小檗碱、苦参碱和氧化苦参碱在各自的质量范围内对色谱峰面积呈现良好的线性关系.本方法准确、灵敏、重复性好,能较全面地评价呋喃苦参黄连素片的质量.We establish an HPLC method for determining four components such as furazolidone,berberine hydrochloride,matrine and oxymatrine in Funan kushen huangliansu tablet. Furazolidone and berberine hydrochloride were detected by the amino chromatographic column: Inertsil NH_2 5 μm 4. 6 × 250 mm. The mobile phase was acetonitrile: anhydrous ethanol: 2% phosphoric acid solution. The UV-vis wavelengths of furazolidone and berberine hydrochloride were set at 365 and 350 nm,respectively. Matrine and Oxymatrine were detected by ODS column: Promosil C_(18)5 μm 4. 6 × 250 mm. The mobile phase was methanol: phosphate buffer solution( 7 ∶ 93). Detection wavelength is 220 nm. The flow rate was 1. 0 mL · min^(-1). The column temperature was 30 ℃. The chromatographic peaks have good resolution and repeatability. Furazolidone,berberine hydrochloride,matrine and oxymatrine have linear relationship in the range of appropriate concentration. The method is accurate,sensitive,reliable,and it has good reproducibility. It can be used for the quality control of Funan kushen huangliansu tablet.
