摘 要:对9株供试乳酸菌产酸产气、生长速率、产酸速率等方面筛选既能用于微生态制剂又能用于青贮饲料发酵剂的优良乳酸菌;并从细菌形态学、生理生化特征和16SrDNA基因序列分析对其进行鉴定;同时进一步研究乳酸菌对温度、盐浓度和强酸强碱的耐受性、有机酸组分和药物敏感性等生物学特性。试验筛选出植物乳杆菌R02(Lactobacillus plantarum)、粪肠球菌R07(Enterococcus faecalis)和戊糖片球菌R09(Pediococcus pentosaceus)3株优良乳酸菌。其中菌株R09生长最快,产酸能力最强,乳酸占总酸含量最高,达95.45%,可耐受0.12g/mL NaCl;R02对温度的耐受性最强,可耐受55℃高温;R07对强酸强碱环境有较强的耐受能力,在pH为3.0~9.5的条件下均可生长。菌株R02、R07、R09均具有生长快、产酸和耐受能力强的优良生物学特性,因此具有应用于微生态制剂和青贮饲料的潜能。The study was conducted to screen the excellent lactic acid bacteria for both silage additive and probiotics based on acid and biogas yield tests,growth rate and rates of acid producing tests,and the isolated strains were identified by morphological observation,physiological and biochemical tests,and sequence analysis of the 16 SrDNA gene.Furthermore,other biological characteristics of excellent lactic acid bacteria,such as salt,acid and alkalis solvent tolerance,temperature resistance,the organic acid of lactic acid bacteria fermenting liquid composition and drug sensitivity were investigated.The result showed that three strains of lactic acid bacteria,R02(Lactobacillus plantarum),R07(Enterococcus faecalis)and R09(Pediococcus pentosaceus)were screened.R09 has the fastest growth rate and the strongest ability of producing acid.The proportion of lactic acid is the highest in liquid cultures of R09,accounting for 95.45% of the total acid content.Additionally,R09 can survive in0.12 g/mL NaCl solution.R02 has the strongest tolerance to temperature,which grows well below55 ℃.R07 has a strong resistance to acid and alkali,which can grow and survive within a wide pH range(3.0 to 9.5).R02,R07 and R09 strains characterized with such as high acids-producing capacity and high tolerance to acid,have a potentiality of using as silage additive and probioties.
