摘 要:目的研究分析AOTF-近红外光谱(Near Infrared Spectrum Instrument,NIRS)在鉴定黄芩中提取物黄芩苷的含量的作用。方法收集10个不同厂家100批黄芩提取物样品,采用AOTF-近红外光仪对样品近红外光谱进行采集,通过HPLC法测定黄芩苷含量,采用偏最小二乘法(PLS)法建立黄芩苷NIR广谱信息及含量间矫正模型。结果建立的最优校正模型R2=0995,RMSEC=0.441,RMSECV=2.260;外部验证,r2=0.987,RMSEP=0.485,平均回收率为100.191%。结论在不同厂家黄芩提取物黄芩苷含量测定中,AOTF-近红外光谱能实现大批量样品检测分析,其具有的快速、准确、无损等优势,可积极推广和应用。Objective To study and analyze the role of AOTF- near infrared spectroscopy (Near Infrared Spectrum Instrument, NIRS)in determination of the content of Baicalin in Scutellaria baicalensis Georgi. Methods Samples of 100 batches of Scutellaria baicalensis extract from 10 different manufacturers were collected,and the near infrared spectra of the samples were collected by AOTF- near infrared spectrometer.Then,the Baicalin content were determined by the HPLC method,and the broad spectrum information and content correction model of Baicalin NIR were established by the Partial least squares (PLS) method.Resuh The optimal calibration model R2=0995, RMSEC=0.441, RMSECV=2.260;and external validation, r2=0.987, RMSEP=0.485, with an average recovery of 100.191%.Conclusion In the determination of Baicalin in Scutellaria baicalensis Georgi extracts from different manufacturers, AOTF- near infrared spectroscopy has the advantages of simple operation, reliable results, high speed, no pollution and so on,and can achieve fast and accurate analysis of large quantities of samples.
