摘 要:目的研究探讨EB病毒微小RNA在鼻咽癌中的表达情况,以便于找到诊断和防治鼻咽癌的生物标志物。方法选取45例鼻咽癌患者和37例鼻咽炎患者为研究对象,借助实时荧光定量PCR技术并以miR-U6作为内参物,对上述疾病病灶中的EB病毒miRNAs(包括EBV-miR-BART4*和EBV-miR-BART18-3P)的表达情况进行研究分析。结果经统计分析可知,EB病毒的两种miRNAs在鼻咽炎组织中的表达数值分别是(28.43±3.16)和(26.73±3.08),而在鼻咽癌组织中的表达数值分别是(40.51±4.82)和(41.66±3.46),可见其在鼻咽癌组织中的表达水平明显高于鼻咽炎组织,两者具有显著的差异性(P<0.01)。结论 EB病毒微小RNAs可通过较显著的表达水平导致鼻咽癌的发病和病灶转移,因此可将其作为鼻咽癌诊断防治中潜在的生物分子标志物。Objective To explore the expression of Epstein-Barr virus micro-RNA in nasopharyngeal carcinoma, and to discover the biomarker for diagnosis and treatment of nasopharyngeal carcinoma. Methods 45 patients with nasopharyngeal car- cinoma and 37 patients with nasopharyngitis were chosen as the research objects. The expression of Epstein-Barr virus micro- RNAs including EBV-miR-BART4 * and EBV-miR-BART18-3P was studied and referred with miR-U6 by means of real-time flu- orescent quantitative PCR. Results Through statistical analysis of experimental data,the expression of Epstein-Barr virus micro- RNAs in nasopharyngitis were respectively (28.43 ± 3.16) and (26.73 ± 3.08 ) ,and of nasopharyngeal carcinoma were respec- tively (40.51 ±4.82) and (41.66 ± 3.46). The expression of Epstein-Barr virus micro-RNAs in nasopharyngeal carcinoma were higher than that of nasopharyngitis,the difference was significant ( P 〈 0. 01 ). Conclusion Epstein-Barr virus micro-RNA can express and result in nasopharyngeal carcinoma at a significantly high level, which means it can be treated as a molecular biomar- ker in the diagnosis ,prevention and treatment of nasopharyngeal carcinoma potentially.
