摘 要:目的观察6-羟基多巴胺(6-OHDA)毁损双侧间脑A11核团对sD大鼠行为表现的影响,探讨此方法建立不安腿综合征(RLS)动物模型的可行性。方法21只健康SD大鼠根据体重随机分成3组,分别为空白组3只,假手术组9只,毁损组9只,麻醉后对大鼠双侧间脑A11做定向注射,空白组不做处理,假手术组注射0.1%维生素c生理盐水,毁损组注射0.2%6-OHDA(舍01%维生素c的生理盐水),1周后录制视频,观察大鼠行为表现,包括站立、爬笼、睡眠次数和时间。视频录制完成后,麻醉后处死取脑,用免疫组化法检测间脑A11核团酪氨酸羟化酶(TH)的表达,统计分析A11毁损率。结果毁损组毁损率明显高于假手术组(P〈0.05),大鼠站立、爬笼、睡眠次数和时间差异均无统计学意义(P〉0.05)。结论6-oHDA定向毁损双侧间脑A11对sD大鼠行为表现无显著影响,是否能作为RLs动物模型还需要进一步实验。Objective To observe the effect of 6-hydroxydopamine ( 6-OHDA ) damaging bilateral diencephalic A11 nucleus on the behavior of SD-rats, and to explore the feasibility of establishing the animal model ofresdess leg syndrome ( RLS ) . Methods 21 healthy SD rats were randomly divided into three groups based on body weight: blank group with 3, sham operation group with 9, damage group with 9. After anesthesia, 6-OHDA was stereotaxic injected into the bilateral diencephalic A11 nucleus.Blank group was not treated, sham operation group was injected with 0.1% vitamins of saline, the damage group injected 0.2% 6-hydroxydopamine ( containing 0.1% vitamins of saline ) . After a week, the video was recorded to observe the behavior of the rats, including upright, climbing cages, sleep times and time. After the video recording was completed, rats were killed to take brain after anesthesia .The expression of tyrosine hydroxylase ( TH ) in diencephalic All nucleus was detected by immunohistochemistry, statistics and analysis of A11 damage rate. Results The damage rate of the damage group was significantly higher than sham operation group ( P〈0.05 ) , but there was no significant difference in the number of rats standing upright, climbing cages, sleeping times and time. Conclusion There are not significant effects of 6-hydroxydopamine directionally damaging bilateral diencephalic A11 nucleus on the behavior of SD-rats, but it requires further experimentation to confirm whether this study can be used as an animal model of RLS.
