摘  要:目的探讨姜黄素联合PDTC体外抗K562细胞的作用。方法 MTT法检测姜黄素单独或联用PDTC后对K562细胞增殖活性的影响;Hoechst33342染色法观察细胞凋亡形态;Western blot法检测Cleaved Caspase 3的表达活性。结果 10μmol·L^(-1)姜黄素联合25μmol·L^(-1)的PDTC后,可显著提高对K562细胞增殖的抑制活性,并显示出诱导细胞凋亡的形态学特征,使细胞Cleaved Caspase 3表达增加。结论姜黄素联合PDTC可增强抗K562细胞作用,机制与诱导K562细胞凋亡作用有关。Objective: To study the effect of curcumin combined with PDTC on anti - leukemia K562 cells in vitro. Methods: MTY assay was used to detect the effect of curcumin alone or in combination with PDTC on the proliferation of K562 cells. The morphology of apoptotic cells was observed by Hoechst33342 staining. The expression activity of Cleaved Caspase 3 was detected by Western blot. Results : After treated with 10 μmol·L^-1 curcumin and 25μmol·L^-1 PDTC, the inhibitory activity on proliferation of K562 cells significantly in- creased, and the morphological characteristics of abduction apoptosis were also proved. The expression of Cleaved Caspase 3 increased. Conclusion: Curcumin combined with PDTC can enhance the anti- K562 cells and its mechanism is related to the abduction apoptosis of K562 cells.
