摘  要:目的为了诠释桑菊饮治疗咳嗽的科学内涵,研究桑菊饮治疗咳嗽的分子机制。方法用辣椒素诱导小鼠咳嗽模型,分别设置正常组,模型组,桑菊饮高、中、低剂量治疗组,记录各组小鼠给药前后的咳嗽次数,采用荧光定量PCR及Western Blot技术对TRPV1的表达量进行测定,采用ELISA法检测小鼠三叉神经节组织中P物质的释放量。结果小鼠被辣椒素刺激后,咳嗽次数明显增加,TRPV1表达上调,与正常组相比差异具有统计学意义(P<0.01或P<0.05),给予不同剂量的桑菊饮水煎剂治疗5 d后,高、中剂量组小鼠咳嗽次数减少,TRPV1表达下调,P物质含量降低,与模型组相比差异具有统计学意义(P<0.01或P<0.05),其中以中剂量组效果最佳,低剂量组效果不明显。结论下调TRPV1的表达而减少小鼠咳嗽次数可能是桑菊饮治疗咳嗽的分子机制之一。Objective To interpret the scientific connotation and determine the molecular mechanism of Sangjuyin decoction for coughing. Methods An experimental cough model cough of mice induced by capsaicin was made. The model mice were divided into a control group, a modle group, and 3 Sangjuyin decoction dosage groups. The times of cough before and after the treatment in each group were recorded, and the expression of TRPV1 was detected with real-time PCR and Western Blot. Results There were apparent increases in the times of cough and the expression of TRPV1 after the mice were irritated by capsaicin, and there was significant difference from the control group (P 〈 0.01 or P 〈 0.05). The times of cough decreased and the TRPV1 expression was down-regulated after receiving the high, and middle doses of Sangjuyin decoction treatment for 5 days, there was a significant difference compared with those of the control group (P 〈 0.01 or P 〈 0.05). Conclusion Decreasing TRPV1 expression to reduce the number of coughs in mice may be one of the molecular mechanisms of Sangjuyin decoction for cough.
