摘　　要：目的研究SLC34A2基因对CD166^+肿瘤干细胞生长的调控作用。方法用RT-PCR和Western-blot检测已验证具有肿瘤干细胞特征的非小细胞肺癌CD166^+细胞和CD166-细胞中SLC34A2的表达水平,然后通过siRNA干扰技术沉默CD166^+细胞中SLC34A2基因,比较SLC34A2基因沉默前后,CD166^+细胞的成球能力和集落形成能力的变化;取沉默或未沉默SLC34A2基因的CD166^+细胞悬液0.5 ml,分别腹腔注射到8-9周的Balb/c小鼠体内(每组6只),比较其体内肿瘤形成能力。结果 SLC34A2在CD166^+细胞的表达量显著高于CD166-细胞(P<0.05);沉默SLC34A2基因后,CD166^+细胞的成球能力有明显下降(P<0.05);同时集落形成的数量也显著减少(P<0.05);接种沉默SLC34A2基因的CD166^+细胞悬液的小鼠体内肿瘤的生长能力受到明显抑制(P<0.05)。结论沉默SLC34A2基因可抑制非小细胞癌CD166^+干细胞的生长和增殖。Objective To explore the regulation effect of SLC34A2 on the growth of CD166^＋ tumor stem cells. Methods The expression levels of SLC34A2 in CD166^＋ cells and CD166^- cells in non-small cell lung cancer（NSCLC） with tumor stem cell characteristics were detected by RT-PCR and Western blot. SLC34A2 in CD166^＋ cells was silenced by siRNA method, the cell growth capacities were detected by sphere-forming and colony formation assays. The 0. 5 ml volume of CD166^＋ cells with or without SLC34A2 knockdown were intraperitoneally injected into 8 - 9-weeks-old Balb/c mice （n = 6 for each group）, the cell growth ability in vivo was measured and compared. Results The expression of SLC34A2 in CD166^＋ cells was significantly higher than that in CD166^- cells （P 〈 0. 05）. The capacity of sphere-forming in CD166^＋ cells was significantly decreased after SLC34A2 silencing（ P 〈 0. 05 ）, and the number of colonies was also significantly decreased（P 〈 0.05）. The growth of tumor in mice injected by CD166^＋ cell suspension with SLC34A2 silencing was significantly inhibited （ P 〈 0. 05）. Conclusion Silencing of SLC34A2 Could inhibit the growth and proliferation of CD166^＋ stem cells in non-small cell carcinoma.
