摘 要:目的:探讨视黄醇代谢相关基因LRAT和RDH12在宫颈鳞癌细胞系Si Ha和Ca Ski中的抗肿瘤作用,及其对细胞增殖、凋亡和侵袭迁移的影响。方法:构建LRAT及RDH12过表达载体并转染宫颈鳞癌细胞系Si Ha和Ca Ski。CCK-8法检测细胞增殖情况,Annexin V-FITC(绿色荧光)和碘化丙啶(PI)(红色荧光)染色转染的细胞,流式细胞仪检测细胞周期、细胞凋亡情况,Transwell小室检测宫颈鳞癌细胞迁移和侵袭能力。结果:转染构建成功的LRAT和RDH12过表达载体后,Si Ha和Ca Ski细胞G_0/G_1期比例增高,S期降低;相较对照组细胞,Si Ha细胞系中过表达目的基因的细胞凋亡率分别增加约11%、10%,Ca Ski细胞系中,两组则均增加约7%;转染LRAT、RDH12组的Si Ha和Ca Ski细胞迁移和侵袭数均少于转染空白质粒组。结论:LRAT及RDH12过表达对宫颈癌细胞系的增殖、凋亡、侵袭、迁移各途径均有抑制作用,提示LRAT及RDH12可能是宫颈癌的抑癌基因。Objective: To investigate the effects of metabolism-related genes LRAT and RDH12 in cervical squamous cell carcinoma cell lines Si Ha and Ca SKi on cell proliferation,apoptosis,cell invasion and migration. Methods: LRAT/RDH12 over expression plasmids were carried to Si Ha and Ca SKi cell lines,respectively.CCK-8 method was used to detect cell proliferation,further through the Annexin V-FITC( green fluorescence) and propyl iodide organism( PI)( red fluorescence) dyeing transfection cells to detect the cell cycle and cell apoptosis.Transwell chamber was used to detect the migration and invation of cervical squamous carcinoma.Results: We successfully constructed LRAT and RDH12 expression vectors and transfected cells.In the G_0/G_1 phase Si Ha and Ca Ski cell lines accounted for a higher proportion than the control group,and in the S phase the proportion was lower.Compared with the control group,the percentage of apoptotic cells in Si Ha cell lines increased by about 11% and 10%,respectively,and in Ca Ski cell lines,both groups were increased by about 7%.In the Si Ha and Ca Ski cells,group transfected with LRAT and RDH12,the number of the migration and invasion was less than the blank plasmid transfected group. Conclusions: By constructing LRAT and RDH12 expression vector and successfully transfection cells,it was suggested that these two genes had inhibitory effects on proliferation,apoptosis,and tumor invasion and migration in cervical cancer cell lines.It is suggested that this gene may be the tumor suppressor gene of cervical cancer.
