摘 要:目的为延长药物的作用时间,降低毒性,增强靶向治疗效果,将青藤碱制成用于肿瘤治疗的长循环脂质体。方法以包封率为指标,采用正交设计法对药品药脂比、胆脂比、聚乙二醇6000(PEG6000)含量、超声时间等因素进行了优化。结果优选工艺为药脂比1∶8,胆脂比1∶3,PEG6000含量0.25%,超声时间15 min。按此工艺进行验证试验3批,测得包封率为61.25%、60.17%、62.33%。结论该法具有工艺稳定、制法简便、重复性好等特点,适于青藤碱长循环脂质体的制备。Objective To prepare the sinomenine long circulating liposomes to prolong the action time of the drug,reduce the toxicity and enhance the targeted therapeutic effects for tumor treatment.Methods With the entrapment rate as the index,the orthogonal design was adopted to optimize the ratio of drug to lipid,the ratio of cholesterol to lipid,PEG 6000 content and ultrasonic time.Results The optimized technology was as follows:the ratio of drug to lipid was 1∶8,the ratio of cholesterol to lipid was 1∶3,the PEG 6000 content was 0.25%,and the ultrasonic time was 15 min.The three batches of validation tests were conducted according to this technology.The detected entrapment rates were 61.25%,60.17% and 62.33% respectively.Conclusion The method has the advantages of stable technology,simple preparation method and good repeatability,so it is suitable for the preparation of sinomenine long circulating liposomes.
