摘  要:目的观察缺血后处理(IPOC)联合水蛭注射液对脑缺血损伤大鼠磷脂酸肌醇-3激酶(PI3K)mRNA及其蛋白表达和脑梗死体积的影响。方法 SD大鼠40只,随机分为假手术组(SO组)、缺血再灌注(MCAO)组、缺血后处理组(IPOC组)、缺血后处理+水蛭注射液后处理组(P-L组)4组。采用线栓法制备大鼠IPOC和MCAO模型,采用TTC染色法计算脑梗死体积,实时荧光定量PCR和Western blot技术检测PI3K mRNA及其蛋白的表达变化。结果大鼠脑缺血再灌注后24h,IPOC组较MCAO组梗死体积明显减小,P-L组脑梗死体积较IPOC组进一步缩小(P<0.05)。SO组的PI3K mRNA及其蛋白的表达量较少,MCAO组的PI3K表达明显增高(P<0.01);IPOC组、P-L组的PI3K表达水平较MCAO组升高(P<0.01);P-L组PI3K表达较IPOC组升高(P<0.05)。结论脑缺血损伤;脑缺血后处理具有神经保护作用,其机制可能与其诱导PI3K的表达有关,水蛭注液可进一步促进其表达而发挥神经保护作用。Objective To observe the effect of leech injection and ischemic postconditioning (IPOC)on the expression of phosphoinositide3 kinase (PI3K)mRNA and protein against ischemia reperfusion (I/R)injury in rats.Methods Forty male SpragueDawly (SD)rats were randomly divided into four groups:sham operation (SO)group,middle cerebral artery occlusion (MCAO)group,IPOC group,and leech injection postconditioning (P L)group.The IPOC and MCAO rat models were established by sutureocclusion method.The volume of cerebral infarction was calculated by 2,3,5 Triphenyltetrazolium chloride (TTC)staining.The expressionof PI3K mRNA and protein was detected by real time fluorescence quantitative polymerase chain reaction (PCR)andwestern blot.Results The infarct volume of brain tissue was significantly lower in IPOC group than that in MCAO group after 24 h followingI/R,which was lower in P L group than that in IPOC group (P 〈0.05).The expression of PI3K mRNA and protein increasedat 24 h following reperfusion in MCAO group (P 〈0.01).The expression of PI3K increased in IPOC group and P L group (P 〈0.05or P 〈0.01),which was higher in P L group than that in IPOC group (P 〈0.05).Conclusion IPOC could protect neurons throughupregulating the expression of PI3K.Leech injection postconditioning could augment the neuroprotection induced by IPOC.
