摘  要:以山杏内种皮为原料,采用超声波辅助萃取法提取磷脂类化合物,并利用高效液相色谱法分离测定含量。以总磷脂得率为评价指标,通过单因素试验和正交试验优化超声波辅助萃取条件。结果表明,以正己烷-乙醇(1∶1,体积比)为提取溶剂,料液比为1∶15(g/mL),超声功率为450 W,超声时间为50 min最优,在此条件下,山杏内种皮总磷脂含量为0.091 2%。高效液相色谱-紫外检测器法测定磷脂含量,采用ZORBAX Rx-SIL型硅胶柱(4.6 mm×250 mm,5μm),流动相为乙腈-甲醇(15∶85,体积比),流速1.0 mL/min,柱温35℃,进样量20μL,测得山杏内种皮中磷脂酰乙醇胺(PE)含量为0.393 4 mg/g。Phospholipids from apricot seed coat were extracted by using ultrasonic assisted extraction and then analyzed by high performance liquid chromatography. The phospholipids yield was served as the index,the extraction technology was investigated by single factor experiment and orthogonal experiment. The results indicated that the optimal extraction parameters were as follows:the extraction solvent was hexane-ethanol(1 ∶ 1,by volume),the material liquid ratio was 1 ∶ 15 (g/mL),the ultrasonic power was 450 W,and the extraction time was 50 min,the total phospholipids content in apricot seed coat was 0.091 2 % under these conditions. Phospholipids were separated on a Silica gel column(ZORBAX Rx-SIL,4.6 mm×250 mm,5 μm),with acetonitrile-methanol(15 ∶ 85,by volume)as the mobile phase at the flow rate of 1.0 mL/min,and detected by a UV detector. The column temperature was 35 ℃,the injection volume was 20 μL,and the phosphatidyl ethanolamine(PE)content in seed coat was 0.393 4 mg/g.
