摘 要:目的观察微小RNA(miRNA,miR)-151-5p和miR-151-3p在甲状腺乳头状癌(PTC)中的表达并探讨其临床意义。方法采用实时定量聚合酶链反应(Real-time PCR)检测200例PTC组织及配对癌旁组织中miR-151-5p和miR-151-3p表达水平,探讨miR-151-5p和miR-151-3p表达与PTC患者临床病理特征的关系。结果miR-151-5p在PTC的相对表达量(8.49±1.18)显著高于癌旁组织(1.91±0.26, P=0.000),有淋巴结转移的PTC组织中miR-151-5p相对表达量(10.35±1.36)显著高于无淋巴结转移者(5.98±1.25,P=0.034)。miR-151-3p在PTC的相对表达量(9.25±1.26)显著高于癌旁组织(2.02±0.31,P=0.000),但PTC转移组(9.41±1.29)与非转移组(9.04±1.23)比较miR-151-3p表达差异无统计学意义(P=0.658)。miR-151-5p高表达与PTC患者性别(P=0.000)、病灶数量(P=0.004)、局部侵犯(P=0.018)、淋巴结转移(P=0.000)和病理分期(P=0.000)显著相关。结论miR-151-5p在PTC组织中表达上调,与淋巴结转移明显相关。Objective To observe the expression of microRNA (miRNA, miR)-151-5p and miR-151-3p in papillary thyroid carcinoma (PTC) and explore its clinical significance.Methods The levels of miR-151-5p and miR-151-3p expression in 200 cases of PTC tissues and paired normal tissues were detected by real-time quantitative polymerase chain reaction (Real-time PCR). The relationship between the expression of miR-151-5p and miR-151-3p and the clinicopathological characteristics of PTC patients was investigated.Results The level of miR-151-5p expression in PTC tissues (8.49±1.18) was significantly higher than that in adjacent normal tissues (1.91±0.26, P=0.000). The level of miR-151-5p expression in PTC patients with lymph node metastasis (10.35±1.36) was significantly higher than that in those without lymph node metastasis (5.98±1.25, P=0.034). The level of miR-151-3p expression in PTC tissues (9.25±1.26) was significantly higher than that in adjacent normal tissues (2.02±0.31, P=0.000). However, there was no significant difference in miR-151-3p expression level between PTC patients with lymph node metastasis (9.41±1.29) and those without lymph node metastasis (9.04±1.23, P=0.658). The high expression of miR-151-5p was significantly correlated with gender (P=0.000), number of lesions (P=0.004), local invasion (P=0.018), lymph node metastasis (P=0.000) and clinical stages (P=0.000) in PTC patients.Conclusion MiR-151-5p is significantly up-regulated in PTC tissues and closely related to lymph node metastasis.
