摘 要:[目的]探讨何首乌酵母发酵提取液对小鼠的肝毒性作用.[方法]取雄性小鼠随机分为正常对照组(蒸馏水10mL/kg)、何首乌酵母发酵提取液大、中、小剂量(5,3,1g/kg)组,末次灌胃给药后禁食1夜,摘取眼球取血,分离血清,测定血清总胆红素、谷丙转氨酶、谷草转氨酶及γ-L-谷氨酰转肽酶等指标水平,取肝匀浆测定超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量.取对数生长期的张氏肝细胞,加入20μL不同质量浓度的何首乌酵母提取液,于72 h后测定抑制率.[结果]何首乌酵母发酵提取液对小鼠肝毒性的实验结果显示,与正常对照组比较,何首乌酵母发酵提取液各剂量组血清总胆红素、谷丙转氨酶、谷草转氨酶及γ-L-谷氨酰转肽酶指标均有不同程度的升高,但差异无统计学意义(P>0.05);与正常对照组比较,何首乌酵母发酵提取液大剂量组SOD活性低、MDA含量增高,但差异均无统计学意义(P>0.05);与正常对照组比较,何首乌酵母发酵提取液小剂量组SOD活性升高、MDA含量降低,两者间差异亦无统计学意义(P>0.05).何首乌酵母发酵提取液对张氏肝细胞毒性的实验结果显示,与正常对照组比较,加入50,100,200 mg/L质量浓度实验组的OD值降低,较加入400,800 mg/L质量浓度实验组的OD值增高,各组间差异均无统计学意义(P>0.05).各组乳酸脱氢酶及SOD,MDA水平间差异亦均无统计学意义(P>0.05).[结论]何首乌酵母发酵提取液对小鼠未见有明显的肝毒性作用,对张氏肝细胞亦未见有毒性作用.OBJECTIVE To study the hepatotoxicity of the fermentation extract of Polygonum multiflorum Thunb in mice.METHODS Male mice were randomly divided into control group(10 mL/kg distilled water),high dose,middle dose and low dose group(5,3 and 1 g/kg fermentation extract of Polygonum multiflorum Thunb).The mice were fasting for one night after the last gavage,and the eyeballs were taken out for the blood.Serum total bilirubin,alanine aminotransferase(ALT),aspartate aminotransferase(AST)andγ-L-glutamyl transpeptidase were determined.The liver homogenate was used for the determination of superoxide dismutase(SOD)activity and malondialdehyde(MDA)content.The logarithmic growth phase Chang liver cells were treated by adding 20μL different mass concentration fermentation extract of Polygonum multiflorum Thunb,and the inhibitory rate was determined after 72 h.RESULTS The hepatotoxicity results showed that as compared with the control group,the concentrations of serum total bilirubin,ALT,AST andγ-L-glutamyl transpeptidase increased in the experimental groups, but there was no statistical significance(P0.05).Compared with the control group,the activity of SOD decreased and the content of MDA increased in the high dose group,but there was no significant difference between the two groups(P0.05).Compared with the control group,the activity of SOD increased and the content of MDA decreased in the low dose group,and there was also no statistical significance(P0.05).The results of cytotoxicity test of fermentation extract of Polygonummulti florum Thunb showed that as compared with the control group,the OD value of the 50,100 and 200 mg/L experimental groups was lower,while it was higher in the 400 and 800 mg/L experimental groups.There was no statistically significant difference between the groups(P0.05).There was no significant difference of LDH,SOD and MDA value between the groups(P 0.05).CONCLUSION The fermentation extract of Polygonum multiflorum Thunb did not have obvious hepatotox
