摘 要:目的:研究氧化白藜芦醇对体外培养人肝癌细胞QGY-7701、SMMC-7721增殖和迁移能力的影响。方法:采用MTT法测定氧化白藜芦醇抑制肝癌细胞QGY-7701、SMMC-7721生长的IC50;采用Transwell法测定经不同浓度氧化白藜芦醇(25μM、50μM和100μM)以及白藜芦醇50μM与细胞共培养24h后细胞的迁移率。结果:MTT实验结果显示,氧化白藜芦醇抑制QGY-7701和SMMC-7721细胞的IC50值分别为100.37±21.97μM和74.39±9.30μM。Transwell实验结果显示,经25μM、50μM、100μM氧化白藜芦醇和白藜芦醇50μM处理24h后,各实验组细胞迁移率分别是74.96±2.85%、60.26±9.25%、38.58±8.69%和46.80±8.43%。且与对照组比较有显著性差异(P<0.05),氧化白藜芦醇浓度越高,细胞迁移率越低。结论:氧化白藜芦醇具有抑制体外培养人肝癌细胞QGY-7701、SMMC-7721的细胞增殖作用;可显著抑制肝癌细胞QGY-7701、SMMC-7721在体外培养条件下的迁移能力,其抑制能力呈浓度依赖性。Objective:To study the effects of Oxyresveratrol on the proliferation and migration of human hepatocellular carcinoma cells:QGY-7701 and SMMC-7721.Methods:Proliferation and growth rate curves have been investigated using MTT;migration rates of the cells have been measured with Transwell assay.Results:MTT tests showed that the cells in the experimental group with the different concentrations of Oxyresveratrol have been significantly inhibited compared with control group.Transwell experiments showed that migration rates of cells in different concentrations of Oxyresveratrol have been significantly decreased compared with the control group.Conclusion:Proliferation and migration rates of human hepatocellular carcinoma cells QGY-7701 and SMMC-7721 can be inhibited by Oxyresveratrol.
