摘  要:目的研究氢气对大鼠脑缺血/再灌注(I/R)损伤脑皮质区线粒体功能的影响机制。方法雄性SD大鼠48只,用线栓法建立大鼠局灶性脑I/R损伤模型。随机将大鼠分为假手术组(Sham组)、脑I/R组(MOD组)、氢气治疗组(H_2组)。再灌注24 h后进行神经功能缺损评分;检测神经细胞活性氧(ROS)生成率,线粒体膜电位(△ψm)水平,通透性转换孔(MPTP)开放度,线粒体肿胀度变化。结果与Sham组相比,MOD组大鼠神经功能缺损评分、MPTP开放度、线粒体肿胀度及ROS生成率增加(P<0.01),△ψm水平降低(P<0.01);与MOD组比较,H_2组大鼠神经功能缺损评分、MPTP开放度、线粒体肿胀度和ROS生成率降低(P<0.01),△ψm水平增加(P<0.01)。结论腹腔注射H_2可保护脑I/R后缺血区线粒体的功能,改善大鼠脑I/R后的神经功能评分。Objective To study the effect of hydrogen on mitochondrial function in cerebral cortex after cerebral ischemia/reperfusion (I/R) injury in rats. Methods 48 male SD rats were randomly divided into sham operation group (sham group) , brain I/R injury group (MOD group) , hydrogen treatment group (H2 group). 24 hours after reperfusion, the neurological deficit scoring was performed. The changes of mitochondrial membrane potential ( △ψm), permeability openness (MPTP), ROS production rate and mitochondrial swelling were detected. Results Compared with the Sham group, neurological deficit score, MPTP openness, mitochondrial swelling degree and ROS production rate were increased in the MOD group (P 〈 0.01 ), △ψm levels were reduced (P 〈 0.01 ). Compared with the MOD group, the neurological deficit score, MPTP openness, mitochondrial swelling degree and ROS production rate were decreased in H2 group (P 〈 0.01), Atlma levels increased (P 〈 0.01 ). Conclusions Simul- taneous intraperitoneal injection of pure hydrogen can reduce the generation of reactive oxygen species, protect the mitochondrial function of neuronal cells in the ischemic region after brain I/R, improvement the rat brain I/R after the neurological scoring.
