摘 要:目的研究探讨皮肤再生医疗技术(moist exposed burn therapy/moist exposed burn ointment,MEBT/MEBO)对大鼠慢性难愈合创面组织中表皮生长因子受体(epidermal growth factor receptor,EGFR)表达的影响。方法将60只SPF级SD雄性大鼠随机分为空白组(12只)、对照组(12只)、模型组(12只)、贝复新组(12只)及MEBO组(12只),其中,空白组大鼠做背部备皮处理后采用生理盐水纱布湿敷;对照组大鼠做深达筋膜层的全层皮肤缺损创面模型后采用生理盐水纱布覆盖治疗;模型组大鼠做慢性难愈合创面模型后采用生理盐水纱布覆盖治疗;贝复新组大鼠做慢性难愈合创面模型后采用贝复新治疗;MEBO组大鼠做慢性难愈合创面模型后采用MEBO治疗。分别于治疗第3、7、14天取各组大鼠相同部位皮肤或创面组织进行HE染色,并用Western blot法检测组织中EGFR的含量,对比观察各组大鼠皮肤或创面组织中EGFR的表达水平及组织病理变化。结果局部处理后第3、7天,除空白组外,其余各组大鼠创面组织HE染色病理切片均显示炎症细胞增多,有新生血管生成;局部处理后第14天,对照组、贝复新组、MEBO组大鼠创面组织中炎症细胞消失,创面再上皮化。局部处理后第3、7、14天,空白组大鼠背部皮肤组织中EGFR表达水平无明显变化,F=0.667,P>0.05,差异无统计学意义;对照组、模型组、贝复新组及MEBO组大鼠创面组织中EGFR表达水平变化均较明显,F值分别为79.546、89.600、63.036、56.628,P均<0.01,差异具有统计学意义,其中对照组、贝复新组及MEBO组大鼠创面组织中EGFR表达水平均呈先升高后降低的趋势,而模型组则呈递增趋势;局部处理后第3、7、14天,各组大鼠皮肤或创面组织中EGFR表达水平组间两两对比,除贝复新组与MEBO组对比,P>0.05,差异无统计学意义外,其余各组组间两两对比,P均<0.05,差异具有统计学意义;局部处理后第3、7、14天,各组大鼠皮肤或创面组�Objective To investigate the effect of Moist exposed burn therapy/Moist exposed burn ointment (MEBT/MEBO) on the expressions of EGFR in chronic refractory surface wounds of rats. Methods 60 Sprague-Dawley (SD) male rats were randomly divided into 5 groups (Blank group, Control group, Model group, rb-bFGF group, and ME- BO group, and each group has 12 rats). Back skin preparation was done on rats in blank group and then covered with wet saline gauze ; full-thickness skin defect wound model that reached the depth of fascia was made on rats in control group and then covered with saline gauze; chronic refracto~~ wound model was made on rats in model group and then covered with sa- line gauze; chronic refractory wound model was made on rats in rb-bFGF group and then covered with rb-bFGF gauze; Chro- nic refractory wound model was made on rats in MEBO Group and then covered with MEBO gauze. On day 3,7 and 14 ',filer treatment, skin or wound tissues in the same part were obtained and HE staining was performed on them and protein expres- sion of EGFR was detected by the Western blotting analysis. The expression levels of EGFR and histopathological changes in skin or wound tissues of the rats in all groups were compared and observed. Results On day 3 and 7 'after local treatment, except blank group, the histopathological slices of the rats in all other groups showed an increase in inflammatory cells and angiogenesis was observed. On day 14 after local treatment, inflammatory cells in control group, rb-bFGF group and MEBO group disappeared and epithelization was found on wounds. On day 3, 7, 14 after local treatment, no significant change in expression level of EGFR in back skin tissues of rats in blank group was found and the difference was statistically not signifi- cant ( F = 0. 667, P 〉 0. 05 ). Significant changes in expression levels of EGFR in back skin tissues of rats in control group, model group, rb-bFGF group, and MEBO group were found and the differences were statistically significant (
